Object:To establish the HPLC fingerprints of the rhizoma et radix Notopterygii from different areas. Qualitative analysis was applied to fourteen batches of volatile oils of Notoptergii rhizoma et radix by gas chromatography mass spectrometry(GC-MS). Comparative studies of in vitro and in vivo anti-inflammatary capacity of the rhizoma et radix Notopterygii from different areas were conducted. In addition, we carried out isolating and identify the chemical constituents of Notopterygium franchetii.Method:High performance liquid chromatography(HPLC) was established to simultaneously determine the content of ferulic acid, bergapten, notopterol and isoimperatorin. Fingerprints was established and analyzed by the software of "Similarity Evaluation System for Chromatographic Fingerprint of TCM". Qualitative analysis were applied to fourteen batches of volatile oils of Notoptergii rhizoma et radix by GC-MS. Peaks were matched in NIST02library. The clearance of hydroxyl was determined by1,10-phenanthroline spectrophotometry assay. The removal of superoxide was evaluated by riboflavine photosensitization method. We have compared the effects of the watersoluable extract of anti-inflammatory acitivity in xylene induced mice ear swelling model and the inhibiton of the acute paw edema of rats by ethanolsoluable extracts of Notoptergii rhizome et radix. And four chemical compounds were separated from N. franchetii by column chromatography.Result:We have successfully simultaneously determined the determination of four compouds by HPLC. The fingerprints of N.incisum which come from Sichuan and Qinghai provinces were established and so did the fingerprints of N. franchetii. The volatile oil of different origins has different antioxidant capacities and the extract of Notopterygii rhizome et radix owns different anti-inflammatory capacities. Four compounds were isolated and identied from the ethanol extract of N. franchetii. They were isoimperatorin, p-hydraoxyphenethyl anisate, nodakenetin and2-Propenoic acid,3-(4-hydroxy-3-methoxyphenyl)-,2-(4-hydroxyphenyl) ethyl ester.Conclusion:The results showed that the content of notopterol and isoimperatorin in N. incisum and N. fanchetii varied notoably. Notopterol was higher in N. incisum, and isoimperatorin was higher in N. fanchetii, while the sum of the two parts were meet the criteria of Chinese pharmacopeia(2010edition). Sichuan province’s fingerprints were similar to Qinghai province’s, however N. fanchetiis fingerprints varied in Sichuan, Gansu and Qinghai provinces. Alpha-pinene, beta-pinene and1,4-cyclohexadiene consists of40%in Banma, Dari, Jiuzhi, Gande in Qinghai province and Dege in Sichuan province, other were more than45%in the total area. Pincipal component analysis and clustering ananlysis classify the two origins. In vitro studies of antioxidant showed that the the clearance of.OH and O2-of different sources of the volatile oil of rhizoma et radix Notoptergii, Minxian and Rangtang had better clearance than others. In vivo study showed that materials from Rangtang, Weiyuan, Xining have anti-inflammatory acitivity in xylene induced mice ear swelling model and sample from Minxian, Xining, Aba and Banma have the inhibiton of the acute paw edema of rats.
