Abstract Objective:To survey the Effect of hydroxybutyl-chitosan thermosensitive membrane composited with nanparticles as CD133antibody carrier on the growth of pig iliac endothelial cells(PIEC).Method:To vaccinate PIEC on bare cobalt-chromium metal stents,rapamycin cobalt-chromium metal stents and hydroxybutyl-chitosan thermosensitive membrane composited with nanparticles as CD133antibody carrier and culture24hours.Then the cell morphology was observed by light microscope.Cell proliferation was measured by Cell Counting kit-8(CCK-8).Vascular endothelial growth factor(VEGF)concentration was determined using enzyme-linked immunosorbent assay(ELISA).Palatelet-derived growth factor(PDGF)concentration was also determined using ELISA.Results:PIEC grew well on thermosensitive membrane,with normal morphology.After24hours culture,the survived PIEC on thermosensitive membrane were significantly more than those on bare metal stents(P<0.05),and on rapamycin-eluting stents(P<0.05).The concentration of VEGF of thermosensitive membrane was significantly more than that on bare metal stents(P<0.05),and on rapamycin-eluting stents(P<0.05).The concentration of PDGF of thermosensitive membrane was significantly less than that on bare metal stents(P<0.05),and on rapamycin-eluting stents(P<0.05).Conclusion:Hydroxybutyl-chitosan thermosensitive membrane composited with nanparticles as CD133antibody carrier can improve the proliferation of vascular endothelial cells. |
