Objective Aim of the present study was to investigate the vascular endothelial growthfactor proteins(VEGFs ) expression′s regulation with atherosclerotic rodents model after cerebralischemia stroke.Methods Rats were randomly divided into control group and model group, and then thelatter, established into the atherosclerotic rats model, were randomly divided into five groups:AS-Sham、AS rodent model sustained ischemia(AS-SI) 0.5h、6h、12h、24h. The primarypathological changes of the aortic root and brain tissues were histologically observed by HEstaining, and analyzed with the level and trend of change of VEGF mRNA by ReverseTranscription Polymerase Chain Reactions (RT-PCR) and VEGF protein byimmunohistochemical three-step method. All datas were analyzed with SPSS13.0. Quantitativedata were given as the mean±SEM( x±s). Statistical analysis was performed by ANOVA,P<0.05.Results 1. General observation: firstly, brain tissues were in edema as difference of the timepoints, and then atrophy and liquefaction necrosis. 2. HE Staining: with the time of ischemia,neuron displayed in atrophy, necrosis or apoptosis. 3. RT-PCR and IHC: To compare each type ofrat model groups with control,and compare four-time-points rat model groups with AS-sham,the formers were significantly upregulated(P<0.05), and changed regularly with the four-timepoints, among which VEGF mRNA and its protein productions had reached the peak in AS-SI 6hgroup. The expression of AS-SI 0.5h VEGF mRNA and protein are up-regulated, AS-SI 6h groupat the top, AS-SI 12、24h lower than 6h, but above the baseline. The positive expression of themodel group is widely distributed in the bilateral cerebral cortex, around the brain microvascularcells and in the hippocampus. The positive particles are not only present in the cytoplasm, butalso around the nucleus a little.Conclusion 1. A solid-state high fat diet could successfully replicate a stable atheroscleroticmodel, and then ligate the bilateral common carotid artery for CIS model. 2. There is asynergistic effect on the regulation of VEGF expression in both of tow diseases, the level ofVEGF expression higher and stronger. 3. There is an advanced mechanism which need to beexplored, and the relationship between VEGF expression and systematic inflammation.Meanwhile, it would provide a theoretical model to evaluate the occurrence、development、treatment and prognosis of cerebral ischemia stroke complicated by atherosclerotic disease.
