Fructus Corni, called Shanzhuyu in Chinese, is dry ripe sarcocarp of the cornaceae plant Cornus officinalis Sieb. et Zucc. Hanzhong in Shaanxi province, Nanyang in Henan province and Hangzhou in Zhenjiang province are the main special producing area. Fructus Corni is a frequently-used traditional Chinese medicine which is used clinically for the treatment of various health problems, such as spontaneous perspiration, night sweating, spermatorrhea, enuresis, vertigo, tinnitus, and impotence. Modern researches on Fructus Corni during the past several decades showed that the cornel total iridoid glycosides are the main active parts in Fructus Corni, with the biological activities of anti-inflammatory, hepatoprotection and immunosuppression, etc. Especially the effective function of neuroprotection, antidiabetic and the complication.Preparation optimization of cornel total iridoid glycosides were performed in our study and a preparation route of cornel total iridoid glycosides was established, which significantly increased the content and recovery of active constituent in final product. Large-scale production of cornel total iridoid glycosides as well as the single compound can make benefit to the quality control and the pharmacology study of Fructus Corni. Meanwhile, a preliminary biological activity research on cornel total iridoid glycosides we prepared by us was carried out.this study consists of following three sections:Section1consist of6chapters:(1Establishing the HPLC determination method of main iridoid glycosides-morroniside, loganin and sweroside in Fructus Corni. They are taken as marker compounds for selecting the most suitable preparation conditions and quality control of Fructus Corni;(2)investing some different pretreatment route of Fructus Corni and the best one was selected. Boil the fresh fruits for10mins and put them out of the water, remove the kernel after cooling, dry them in the oven at temperatures of40℃;(3)enzyme hydrolysis method was carried out for improving extraction efficiency of cornel total iridoid glycosides. Comparative experiment between several kind of enzyme showed that amygdalase was the best, the contents of active constituents in extract after enzyme hydrolysis by amygdalase were10%higher than the common method;(4)combing overall desirability and central composite design-response surface methodology, extraction conditions of cornel total iridoid glycosides were optimized. The optimal extraction parameters:Alcohol density (X1):25%-27%, Ratio of liquid to material (X2):12-16BV, Extraction time(X3):30-40mins;(5)combing overall desirability and central composite design-response surface methodology, purification conditions of cornel total iridoid glycosides were optimized. The optimal purification parameters:alcohol density (X1):48%-55%, elution volume (X2):8-9BV, sample volume (X3):40-50mg/g;(6)other related research of Fructus Corni were investigated, including storage of fresh crude drugs, other part of C. officinalis Sieb. et Zucc. plant which is not the Medicinal parts and identification of counterfeit of Fructus Corni.Section2:A preliminary biological activity research on cornel total iridoid glycosides we prepared was carried out. Anti-hypoxia experiment under normal pressure showed that cornel total iridoid glycosides of small (25mg/kg/day), medium (50mg/kg/day) and large-dosage group (100mg/kg/day) could prolong the survival time of hypoxia mice with statistically significant differences compared with normal saline group (P<0.01). cornel total iridoid glycosides of medium (50mg/kg/day) and large-dosage group (100mg/kg/day) indicated statistically significant differences compared with positive drug group (Nimodipine Tablets and Ginkgo Leaves Tablets)(P<0.01). Cornel total iridoid glycosides improving brain hypoxia tolerance of mice in a dose-effect relationship.
