Objective: To central composite design response surface methodology(CCD-RSM) Monomeric flavonoids investigated in vitro drug combination anti-tumor and anti-anoxic injury effects, to find the best combination of flavonoid monomer ratio of drugs to improve the effectiveness of the drug in the clinical treatment.Methods: Using MTT method(MTT colorimetric method) estimates the cell viability, using biochemical assay for intracellular malondialdehyde(MDA) and superoxide dismutase(SOD) levels of testing. With the Design- Expert software level of multiple factors on flavonoids monomer compatibility Design of experiment, the experimental results are mathematical models to predict.According to the binomial fitting equation plotted factors affecting trends in the indicators dimensional and two-dimensional response surface contour map,select the best ratio and experimental verification.Results: Anti-oxidation experiments with hydrogen peroxide 2 hours after injury Schwann cells(RSC) compared to administration groups can maintain high cell viability. The 4- factor ratio of anti-oxidative damage effect are(Luteolin237.5μmol/l+quercetin112.5μmol/l+kaempferide112.5μmol/l+Isor hamnetin 87.5μmol/l),with the survival rate of(69.42%±5.65).In Anti-tumor experiments,the best 4-factors of antitumor effect to A549 is(Luteolin 324.5μmol/l+quercetin201.12μmol/l+kaempferide175.07μmol/l+Isorhamneti n 324.30μmol/l),with the inhibition rate of(63.60%±4.96)and the best 4-factors of antitumor effect to MCF-7 is(Luteolin324μmol/l+quercetin400μmol/l+kaempferide325μmol/l+Isorhamnetin325μmol/l),with the inhibition rate of(70.14%±5.21).Conclusion: The CCD-RSM for different monomer antioxidant flavonoids multivariate multilevel combination of drugs, antineoplastic experimental design, based on the results of the program and then predict the optimum concentration ratio, by verifying finally get the most excellent combination of matching results. This method is simple, good predictability, improved experimental precision experiments in the field of development for the pharmacy to provide new methods and ideas. |
