Background Aquaporin (AQP) water channels are expressed in manydifferent tumor cell types in human. New evidence for the involvement of AQPs incell migration and proliferation adds AQPs to an expanding list of effectors in tumorbiology.Aims The aim of this study was to investigate the role of aquaporin-4(AQP4)on migration and proliferation of human colon cancer cell.Methods AQP4expression was determined by Western blot after vascularendothelial growth factor(VEGF)or AQP4-siRNA treatment in human colon cancercell line LoVo,and the cell migration and proliferation were tested by cell scratchassay and CCK-8proliferation assay.Result VEGF up-regulated AQP4expression of LoVo cell in a time-orconcentration-dependent manner, and increased cell migration.When AQP4wasknocked down by AQP4-siRNA, cell migration decreased significantly. Comparedwith the VEGF-treated cells,the migration of combined AQP4-siRNA and VEGFtreated cell decreased. AQP4-siRNA or VEGF had no significant effect on cellproliferation.Conclusion AQP4may promote the migration of human colon cancer cell.This provide evidence for a novel role of AQP4in tumor growth and spread of humancolon cancer....
