| Objective:1.Human breast cancer cells MDA-MB-231are subcutaneously vaccinated in nude mice in order to establish breast cancer transplantable tumor model of nude mice.2.To prove the effect of Zhuang Gu Zhen Tong(ZGZT) capsule preventing the happening of the Bone tumors caused by breast cancer in the experimental Angle, and to explore,on the molecular level, the mechanism of the effects in the aspect of tumor adhesion,infiltration and metastasis, to providing a powerful pharmacodynamics experimental evidence for ZGZT capsule preventing and treating the bone tumors caused by breast cancer.Methods:32BALB/c nude mice, the weight approximately17.27±0.27g. SPF, female. The suspension of breast cancer cells was uesd to vaccinate in subcutaneousness of nude mice under their shoulder, in order to build Nude mice breast cancer transplant tumor model, and Nude mice were randomly divided into four groups,8nude mice per group, group A:model group (model control group), group B:prevent group (ZGZT capsule prevent group), group C:treatment group (ZGZT capsule treatment group), group D:control group (Disodium Clodronate Capsules control group).The nude mice of group B were feed with ZGZT capsules concentrated liquor450mg/kg after the day they were vaccinated. The Nude mice of A, C and D group were treated separately with different treatment when the cubage of their breast cancer transplant tumor become0.5CM’(about two weeks), group A with0.2ml distilled water, group C with ZGZT capsule concentrated liquor450mg/kg,Group D with Disodium Clodronate Capsules2g/kg. All of the treatment were implemented2times a day, and lasts30days. The transplant tumor cubages were measured each5days. In30days,We would put the four groups of nude mice to death, separate transplant tumor tissues, note the weight of transplant tumor respectively, and calculate the tumor inhibition rate, and would check out each groups’difference of integrin-α νβ3(ITG-α ν β3) and bone sialoprotein (BSP) with Immunohistochemical method.Results:1. MDA-MB-231cells were injected subcutaneously into nude mice two times in a row,10days later, all the groups of nude mice were tumorigenic.2. After different intervention. Comparing each groups of nude mice in general, prevention group was the best, the treatment group and the control group were in general, model group was the worst.3.The tumor inhibition rate of each groups:model group and the treated group had significant difference P<0.01, no statistically significant difference between the treatment group and the control group P>0.05,which compared with prevent group had statistically significant difference P<0.05.4. Transplanted tumor tissues of each group were observated by light microscopy:the tumor cells of the model group arranged compact and even overlap, and the structure was clear, and had tumor nest. Other groups’were pyknosis, and had deformative contour, at the same time,they appeared various degree of apoptosis, Transplanted tumor tissue of the prevention group appeared apoptotic body.5. The ITG-ανβ3and BSP in transplanted tumor tissue of model group was higher than that of the prevention group, the treatment group and the control group (P<0.05), Compared with the prevention group, the treatment group and the control group had statistical difference (P<0.05),the treatment group and the control group showed no significant difference (P>0.05).Conclusion:Zhuang Gu Zhen Tong capsule has a preventive and therapeutic effect on bone tumor caused by breast cancer.and can significantly inhibit the expression of ITG-α ν β3and BSP in breast cancer transplanted tumor tissue, and can inhibit the adhesion and invasion of breast cancer transplant tumor cells by regulating the above protein mediated pathway, so as to achieve function of preventing bone metastasis of breast cancer. |
