| Objective: To investigate the effects of Prevotella intermedia(P.intermedia) andPorphyromonas gingivalis(P.gingivalis) in biofilms by extract extracellular andintracellular proteins of Streptococcus sanguis(S.sanguis).Methods:1.Extract extracellular proteins of S.sanguis by high speed centrifugation,usingorganic solvent.2.By ultrasonic, ammonium sulfate salting out,Sephadex G-25column desaltingthrough dialysis to extract intracellular proteins.3.Observe the effects of P.intermedia and P.gingivalis which roled of theintracellular proteins and extracellular proteins of S.sanguis.4.Observe the effects of P.intermedia and P.gingivalis on biofilm which roled ofthe intracellular proteins and extracellular proteins of S.sanguis.Results:1.The growth of P.intermedia and P.gingivalis is significantly inhibition effectwhich roled of intracellular proteins of S.sanguis.The MIC of intracellular proteins onmixed P.intermedia and P.gingivalis is0.125g/L;there are no significant effect ofP.intermedia and P.gingivalis which roled of extracellular proteins of S.sanguis.2.Intracellular proteins of S.sanguis role in the biofilms of P.intermedia andP.gingivalis mixed,the biofilms activity was decreased,compared with the control grouphave significant differences(P <0.05);however, which roled of the extracellular proteins ofS.sanguis,the difference was not statistically significant which on the activity of biofilmcompared with the control group.Conclusions:1.P.intermedia,P.gingivalis and the mixed bacteria in biofilms which roled ofintracellular proteins of S.sanguis have significantly inhibition effect.2.There are no significantly inhibition effect when the extracellular proteinsrole of the P.intermedia,P.gingivalis and the mixed bacteria in biofilms. |
