Objective:This study is to establish the model of influenza virus pneumoniainfection of infant mice using the vivo experimental techniques, study the roleof prevention and treatment of the extracts of Yupingfengsan and Yinqiaosan fiveflavors to influenza virus pneumonia infection of infant mice, explore its immunemechanism of anti-fluvirus pneumonia,provide an experimental basis for theclinical prevention and treatment of influenza virus.Materials and methods:1. establish the model of influenza virus pneumonia infection of infant mice,verify the model of influenza virus pneumonia of infant mice through breathing,fur, diets, change of weight, appearance of lung tissue, lung pathology in lightmicroscope and the lung index.2. By immunohistochemical methods, detect the IL-2protein expression of lungtissue mice at different time points by administration of the extracts ofYupingfengsan and Yinqiaosan five flavors. explore its immune mechanism ofanti-fluvirus pneumonia.3. Using influenza A virus to infect mice. combine with its general state, lunghistopathology, morphology, lung index and so on, observe the role of preventionand treatment of the extracts of Yupingfengsan and Yinqiaosan five flavors toIV pneumonia in mice.Results:1.The normal group remained a good state, the model group have a change in termsof breathing,fur,diet,lung histopathology and appearance.The lung index of Thetwo group mice significant difference(P<0.05).2.Lung index Results:the lung index of model group significant difference compared with normal group,(P<0.05).with the exception of the shuangbiao lowgroup,other groups have significant difference compared with model group(P <0.05).The model group significant difference compared with the ribavirin group.3.Spleen index results:the spleen index of model mice increased but have nosignificant difference compared with normal group(P=0.635>0.05);shuangbiaogroups and the ribavirin group of mice infected with influenzathe virus have no significant effect(P>0.05),spleen index of mice in each group have nostatistical significance.4.Thymus index results:the thymus index of the model group was significantly reduced compared with normal group(P=0.019<0.05);the thymic index of theshuangbiao groups and the ribavirin group infected with the influenza virusdecreased but not significant improvement(P>0.05);the thymus index of theshuangbiao high group increased significantly compared with model group,(P=0.034<0.05).5.IL-2protein expression of Lung tissue:each inter-group have no statistical significance the first day(p=0.986>0.05);each group have a small amount of IL-2secretion. the model group have no significance compared with normal group(p=0.725>0.05).IL-2protein expression of the shuangbiao group and the ribavirin group begin to increase but have no nosignificant difference compared with model group(p>0.05). each inter-group have statistical significance the third day(p=0.000<0.05);IL-2protein expression of each Lung tissue infected by influenza virus group increased(p<0.05)compared with thenormal group; IL-2protein expression of the shuangbiao group and the ribavirin group improve significantly compared with the model group(p<0.05),it has no significant difference between the high dose of shuangbiao group and the ribavirin group(p=0.912>0.05),it has no significant difference between the middle dose and the low dose of shuangbiao group(p=0.494>0.05).each inter-group have statistical significance the fifth day(p=0.000<0.05).modelgroup have significantly difference compared with the normal group(p<0.05);IL-2protein expression of the shuangbiao group and the ribavirin group increase significantly and have a significant difference Compared with model g roup (p<0.05),it has no significant difference between the high dose of shuangbiao group and the ribavirin group(p=0.868>0.05),it has no significant difference between the middle dose and the low dose of shuangbiao group(p=0.497>0.05).Conclusion:1.Mouse with influenza virus to adapt to the strains intranasal influenza virus infection in infant mice can be successfully copied mouse pneumonia model2.The different doses of extracts of Yupingfengsan and Yinqiaosan five flavors alleviate the inflammation of the lungs caused by influenza virus infection and pulmonary pathological damage etc.. The shuangbiao high group is the best.3.The spleen index of mice infected with influenza virus of each shuangbiaodose group have no significant effect. may be the body’s anti-influenza virus infection is mainly caused by the immune response of the lymphatic system, and not cause obvious change internal structure of the spleen yet.The thymus index of mice infected with influenza virus of each shuangbiaodose group reduced but no significant improvement.that prompts each shuangbiao dosegroup could inhibit the shrinking of the mouse thymus.4.Each shuangbiao dose group could reduce the inflammation of the lung injury in mice,promote the expression of IL-2,effectively prevent the virus from immune injury of the lung tissue.
