Focus on investigating the roasting process mechanism of Radix Vladimiria, putting forward a hypothesis:the effect changing between Vladimiriae Radix before being roasted and after being roasted is related with the differences in chemical composition and gastrointestinal absorption of the primary efficacy composition. This paper has investigated the chemical composition and gastrointestinal absorption dynamics of Vladimiriae Radix before and after being roasted scientifically.In the first part of the paper, through the establishment of HPLC chromatographic conditions and preparation methods of samples and so on, under a premise condition by quality control samples intermittent injection, principal component analysis (PCA) and partial least square-discriminant analysis (PLS-DA) were used to analysis the dataset. Found that Vladimiriae Radix before and after being roasted can be separated from the perspective of chemical composition and the amount change of peak6, peakl8, peak4, peek5, peakl3, peak47, peak48is closely related to the main differences. Peak47is Costunolide and peak48is Dehydrocostus lactone. The conclusion of this part are consistent with the hypothesis that the effect changing is related with the differences in chemical composition.In the second part of the paper, the method of in sute perfusion in rats was adopted, and the HPLC methods of the primary efficacy composition were estabished. Found that Costunolide and Dehydrocostus lactone were absorbed heavily by stomach, small intestine and colon, meanwhile Costunolide was degraded in the stomach. The hourly absorption percentage of Costunolide of Vladimiriae Radix before being roasted were46.57%,72.84%,35.97%and the hourly absorption percentage of Dehydrocostus lactone of Vladimiriae Radix before being roasted were34.08%,75.93%,43.01%. The hourly absorption percentage of Costunolide of Vladimiriae Radix after being roasted were42.13%,52.91%,43.70%and the hourly absorption percentage of Dehydrocostus lactone of Vladimiriae Radix after being roasted were31.81%,68.83%,48.36%. Statistical analysis showed that the hourly absorption percentage of Costunolide and Dehydrocostus lactone of Vladimiriae Radix before being roasted were bigger than those of Vladimiriae Radix after being roasted in stomach and small intestine (P<0.05or0.01). The hourly absorption percentage of Costunolide and Dehydrocostus lactone of Vladimiriae Radix after being roasted were bigger than those of Vladimiriae Radix before being roasted in colon (P<0.01). The conclusion of this part are consistent with the hypothesis that the effect changing is related with the differences in gastrointestinal absorption.The innovate points in this article are:metabolomics platform and gastrointestinal tract absorption which is the first part of ADME were used to study the roasting process mechanism of Radix Vladimiria. The study can expound the roasting process mechanism of Radix Vladimiria from the perspective of chemical composition and gastrointestinal tract absorption and provide a new way to research the mechanism of Traditional Chinese Medicine processing.
