| Objective:Investigation of Crude Radix Polygalae (远志)and Magnolia(厚朴)and compatibility proportions with gastrointestinal electrophysiology and the influence of gastrointestinal smooth muscle from the body and its mechanism.Methods:①Using·the self-made silver needle three electrode, using biological function BL-420E+ experimental system compatibility with Crude Radix Polygalae and Magnolia and compatibility to examine magnoliae rat gastrointestinal electricity slow-wave influence;②Adopt BL-420+ biological function experiment system,to observe the effect of gastrointestinal smooth muscle from the body rabbit and rat;and investigation of acetylcholine (Ach), histamine (HA), barium chloride (BaCl2) tools such as gastrointestinal smooth muscle caused by drugs excitability intervention mechanism.Results:(1) Gastrointestinal electric experimental results:①Compared with blank group, Crude Radix Polygalae can Significantly reduce rat gastrointestinal electricity slow wave amplitude (P<0.05),and have a lower stomach electricity slow wave frequency trend (P>0.05), with compatibility 1:1 could significantly reduce rat intestinal electricity slow wave amplitude (P<0.05); with compatibility 1:2,1:3 enhanced intestinal electricity slow wave amplitude trend (P>0.05).②Compared with Crude Radix Polygalae group,with compatibility 1:2 could significantly enhanced gastric electrical slow wave amplitude (P<0.01),with compatibility 1:3 could significantly enhanced slow wave amplitude and frequency (P<0.01,P< 0.05); Magnolia group could enhanced slow wave amplitude (P<0.05).③Each to rat intestinal electricity slow wave frequency has no significant influence. (2) Vitro Smooth muscle from the rabbit body experimental results:①Crude Radix Polygalae has significant inhibitory effect on intestinal smooth muscle from the body(Inhibition rate for 59.21%); Magnolia group,with compatibility 1:3 have the inhibition rate of intestinal muscle very significantly lower than raw vision (P< 0.01);with compatibility 1:1,1:2 also could inhibition of intestinal muscle excited, inhibition rate significantly higher than that of Magnolia group (P<0.01).②Crude Radix Polygalae could Significantly inhibited Intestinal muscle excitability vitro contraction caused by Ach(Inhibition rate for 21.00%), inhibition rate significantly higher than that of Magnolia group and with compatibility 1:3 (P< 0.05, P< 0.01);Compatibility 1:1.1:2 group can also against intestinal muscle excited contraction caused by Ach,the inhibition rate is lower than the trend of Crude Radix Polygalae (P>0.05).③Crude Radix Polygalae could Significantly inhibited intestinal muscle excitability vitro contraction caused by HA (Inhibition rate for 35.77 %);Compatibility 1:3 group's inhibition rate for (9.91%) significantly below Crude Radix Polygalae group (P<0.05);Compatibility 1:1,1:2 group's inhibition rate for (27.59%.18.03%) significantly higher than Magnolia group (P<0.01).④Crude Radix Polygalae could Significantly inhibited Intestinal muscle excitability vitro contraction caused by BaCl2 (Inhibition rate for 26.04%), Magnolia group. Compatibility 1:2,1:3group's inhibition rate for(3.05%,10.91%,5.88%)significantly higher than Crude Radix Polygalae group (P<0.01);Compatibility 1:1 group's inhibition rate for(17.70%), significantly higher than Magnolia group (P<0.01) (3) Stomach muscle from the rat body experimental results:①Crude Radix Polygalae,Compatibility 1:1.1:2. 1:3group can Significantly increased Stomach muscle from the rat body average amplitude, but can reduce shrinkage frequency Magnolia group can increased Stomach muscle shrinkage frequency and average amplitude, Compared with Crude Radix Polygalae group, has an extremely significant difference (P<0.01); Compatibility with Compatibility 1:2. 1:3group significant differences between the groups (P<0.01).②rude Radix Polygalae,Compatibility 1:1.1:3group could Significantly inhibited Stomach muscle excitability vitro contraction caused by Ach, inhibition rate significantly greater than Magnolia group (P<0.01);Magnolia group. Compatibility 1:2 group also can inhibit its amplitude.③Crude Radix Polygalae could Significantly inhibited Stomach muscle excitability vitro contraction caused by BaCl2,and causes the Ach-induced contraction; Magnolia group,Compatibility1:1,1:2,1:3 group Can significantly reduce its amplitude, compared with Crude Radix Polygalae have extremely significant difference (P< 0.01)Conclusions:①Crude Radix Polygalae can significantly inhibit the rat's slow wave amplitude both in antrum and duodenum, the same inhibition results occurred in the isolated rabbit duodenal smooth muscle contraction amplitude and frequency, which showed a clear dose-effect relationship, it also can reduce the frequency of gastric smooth muscle contraction,to against excitement of gastrointestinal muscles caused by Ach and HA.②Magnolia can significantly enhance the rat's intestinal slow wave amplitude, but it show no influence both in normal and given positive drugs such as Ach,HA,BaCl2 intestinal muscle in vitro; it can increase the contraction of isolated gastric smooth muscle amplitude and the contraction frequency simultaneously, and partially reduce the contraction of gastric smooth muscle induced by Ach and BaCl2.③The compatibility of Crude Radix Polygalae with Magnolia at the ratio of 1:1,1:2,1:3 all can increase the rat's slow wave amplitude both in antrum and duodenum, while the 1:2 andl:3 can also increase gastric slow wave frequency, the more Magnolia, the compatibility of Crude Radix Polygalae with Magnolia showed a lower inhibition to intestinal smooth muscle. Based on our research, we speculate the possible mechanism of the relief caused by the compatibility of Crude Radix Polygalae with Magnolia should include intervening cholinergic Ach and HA related receptor and reducing the direct inhibition to gastrointestinal smooth muscle in vitro caused by Crude Radix Polygalae.This study suggests:The compatibility of Crude Radix Polygalae with Magnolia can relieve gastrointestinal motility disorder caused by Crude Radix Polygalae may through influencing the gastrointestinal electrical activity and mediating M receptors,H1 receptors and other related receptors and local hormones.Compared with compatibility 1:1 group. 1:2,1:3group can adjust gastrointestinal smooth muscle and obviously against Crude Radix Polygalae. |
