| [Objective] We adopt the cultivation of human umbilical vein endothelial cells(HUVEC)to study the effect and mechanism of yiqijieduhuoxue formula on the vascular therapy targeting colon cancer. It is not only conducive to the development of the theory and practice of traditional Chinese medicine "targeted treatment of cancer", but also provide a basis for the development of future drug.[Methods]In this study, we adopt the cultivation of HUVEC and extract serum without drug, serum containing yiqijieduhuoxue formula and serum containing5-FU from the SD rats. At the same time, HUVEC are divided into drug-free serum group (referred to as the control group), traditional Chinese medicine serum low-dose intervention group (referred to as TCM low-dose group), traditional Chinese medicine serum medium-dose intervention group (referred to as TCM medium-dose group), traditional Chinese medicine serum high-dose intervention group (referred to as TCM high-dose group),5-FU serum intervention group (5-FU group). We observe serum containing yiqi jieduhuoxue formula on HUVEC at different time (24h,48h,72h), the activity of endothelial cell proliferation, and immunohistochemical method is used to detect the expression of MMP-9and VEGF.[Result]1. The influence of HUVEC proliferation:When drugs stimulate24h, it shows the inhibition of HUVEC proliferation. With the increase of serum concentrations of yiqijieduhuoxue formula, the proliferation activity of HUVEC gradually decreases, showing a dose-dependent, at24h,48h,72h shows a good effect;5-FU group also significantly inhibits proliferation activity of HUVEC. Each treatment group and control group are significant differences(P<0.05or P<0.01). At48h,72h, TCM medium-dose group, TCM high-dose group with the the5-FU group are compared with a significant difference(P<0.05or P<0.01).2. The influence of VEGF and MMP-9expression:The positive cell optical density values of the control group are higher. The positive cell optical density values of each treatment group are significantly lower than the control group(P<0.05or P<0.01). The positive cell optical density values of TCM medium-dose group and high-dose group with the5-FU group are more significantly lower(P<0.05or P<0.01).This shows that each treatment group are able to reduce the expression VEGF and MMP-9and TCM medium-dose group and high-dose group are the most obvious.[Conclusions]1.The influence of HUVEC proliferation:The yiqijieduhuoxue formula and5-FU can inhibit the proliferation of HUVEC, and the higher dose of TCM, the more obvious inhibition.2. The influence of VEGF and MMP-9expression:The yiqijieduhuoxue formula and5-FU can reduce the influence of VEGF and MMP-9expression on HUVEC, and the higher dose of TCM, the more obvious inhibition.The yiqijieduhuoxue formula can inhibit the proliferation of HUVEC and the mechanism may reduce the VEGF and MMP-9expression on HUVEC. |
