Objective:Based on the pioneering chloasma pathological physiology, this paper sets up the in vitro model which treat chloasma with broad development prospects of pharmacodynamics by studying a-MSH cells of mouse B16melanoma in vitro effect of cell biology, specifically by figuring out the dose-and time-effect of a-MSH affected mice B16melanoma cell proliferation, tyrosinase activity and melanin generate Through experimental research, it is found that Chinese herbal medicine licorice flavonoids is likely to produce whitening effect on chloasma with the best dose.Methods:1. a-MSH experiment:laboratory culture of mouse B16melanoma cells, the results were compared with blank group, with1,10,100,1000ng/ml four concentration of a-MSH as culture medium, the cell to intervene. MTT method, NaOH cracking process and dopamine oxidation method are used to detect melanoma cells in cell proliferation, melanoma cell melanin synthesis and tyrosinase activity.2. Licorice flavonoids best dose within the scope of drug toxicity experiment:configuration of licorice flavonoids four different (62.5,125,250,500ug/ml) of concentration on a-MSH optimum cultivation model, using the same method to detect melanoma cell proliferation, melanoma cell melanin synthesis and tyrosinase activity.Results:1. a-MSH experiment:four doses of cells in the experiment of synthesis, cell proliferation, black have better inhibition of tyrosinase activity is10ng/ml (P<0.05), and48h after training to achieve the best value.2. Licorice flavonoids best dose within the scope of drug toxicity experiment:licorice flavonoids four doses of250ug/ml index effect is best,500ug/ml due to cytotoxicity, not measured.Conclusion:A-MSH can affect mice B16melanoma cell proliferation, tyrosinase activity and melanin formation. Licorice flavonoids on tyrosinase activities have very good inhibitory effect, and can be used for chloasma disease treatment.
