| Objective:To observe the effect of Bushen lluoxue Formula for IL-1, IL-6, TNF-a and the moisture content of articular cartilage in osteoarthritis (OA) through a stable osteoarthritis modeling, to compare the effect Bushen Huoxue Formula with other therapy and to probe into the possible mechanism of the formula for osteoarthritis (OA).Methods:Build stable rats model of osteoarthritis through intra-articular injection of papain. The rats were divided into6groups and feed respectively with Bushen lluoxue Formula, Meloxicam and distilled water. After28days’ feeding, the articular cartilages and blood were taken for test. Test the concentration of IL-1, IL-6and TNF-a in serum and the moisture content of ar t icular cart ilages.In clinic,70patients with osteoarthritis were divided into two groups randomly. Each group takes Bushen Huoxue Formula and Meloxicam respectively. The treatment went on for4weeks. The scores of the symptoms were checked before and after the treatment. The effect of Bushen lluoxue Formula and Meloxicam was compared through the scores.Results:In experiment, the concentration of IL-1β, IL-6and TNF-a in serum of rats model of osteoarthritis were higher than that in normal rats(P<0.05). the concentration of IL-1β, IL-6and TNF-αx in serum of rats treated with high dose and middle dose of Bushen Huoxue Formula an Meloxicam were lower than that in the rats models (P<O.05). There was no significant difference between high dose and middle dose of Bushen Huoxue Formula (P>0.05). Compared with that in the normal rats, the moisture content of articular cartilage in rats models of ostcoarthritis was significantly higher (P<0.05). The moisture content of articular cartilage in high dose and low close of Bushen Huoxue Formula group was significantly lower than that in rats models, Meloxicam group and low close of Bushen Huoxeu Formula group(P<0.05).In clinic, Bushen Huoxue Formula and Meloxicam were both effective for knee osteoarthritis. The control and excenllent rate of Bushen Huoxue Formula was65.63%, the totally effective rate was100%. The control and cxcenllent rate of Meloxicam was66.67%, the totally effective rate was93.33%. There was significant difference between the two groups.Conclusions:1. Intra-articular injection can be a good way to build stable rats models of osteoarthritis.2. Kidney defficiency and blood stasis were the main pathogenesis of osteoarthritis in the theory of TCM.3. Bushen Huoxue Formula can reduce the serum IL-1β, IL-6and TNF-α in rats models and the effect of the formula was better than that of Meloxicam.4. In improving the clinical symptoms of osteoarthritis, Bushen lluoxue Formula was more effective than Meloxicam. |
