Objective:Irritable bowel syndrome is a kind of abdominal pain,abdominal distension,bowel habit change and mucous stool of clinical syndrome,is a typical psychosomatic disease.IBS patients with mental disorder or mental abnormality. Acupuncture can not only alleviate gastrointestinal symptoms, also can adjust the person’s mood and psychology.So,we will choose the baihui point and zusanli point to observe the effects of electroacupunture(EA) on the irritable bowel syndrome model rats’visceral sensitivity and emotional psychology behavior change. Combination of NK1receptor expression in colon tissue and calcitonin gene related peptide mRNA expression within the thalamus,5-HT in colon tissue and hippocampus. Disscuss the parts of the mechanism of IBS with emotional psychological disorder in acupuncture treatment.Methods:To establish wista young rats as the model of irritable bowel syndrome. We evaluated the praxiology of the model rats after the preparation. Rats models with IBS were divided into blank control group,model group,model bound group,EA "Baihui and Zusanli"group(Hereinafter referred to as the EA group). The blank control group and the model group were done nothing,the model bound group was bounded but not given EA,the EA group was started to be given EA. To rat on visceral sensitivity evaluation,open-field test,elevated plus maze,after the end of treatment. After the observation,measure the expression of CGRPmRNA, NK1mRNA, and5-HT level.Results:1After the model preparation:(1)The estimate of the visceral sensitivity:compared with the blank control group, the value of the model group rat belly raising and back hunch-up decreased(P<0.05).(2)The open-flied test:compared with the blank control group,the value of the model group level activities and vertical activities decreaded obviously(P<0.01).(3)The test of elevated plus-maze:compared with the blank control group,the OT%of the model group decreased significantly(P>0.05),the OE%of the model group increased(P<0.05).2After the treatment:(1)The estimate of the visceral sensitivity:compared with the blank control group,the value of the model group rat belly raising and back hunch-up decreased(P<0.05).compared with the model group,the value of the EA group rat belly raising and back hunch-up increaded (P<0.01,P<0.05);compared with the model bound group,the value of EA group rat belly raising and back hunch-up incread (P<0.01,P<0.05).(2)The open-flied test:compared with the blank control group,the value of the model group and model bound group rat level activities and vertical activities decreaded obviously(P<0.01,P<0.05)compard with the model group,the value of EA group rat level activities and vertical activities increased obviously(P<0.01),compared with the model bound group the value of the EA group rat level activities and vertical activities increaded(P<0.05).(3)The test of elevated plus-maze:compared with the blank control group,the OE%and OT%of the model group decreased significantly(P<0.05);compared with the model group,the OE%and OT%of the EA group increased significantly((P<0.01,P<0.05).(4)NK1mRNA expression:Compared with the blank control group,the expression of NK1mRNA was in model group and model bound group;compared with the model group,the expression of NK1mRNA was decreased in EA group(P<0.05).(5)CGRP mRNA expression:Compared with the blank control group,the expression of CGRPmRNA was enhanced significantly in model group and model bound group;compared with the model group,the expression of CGRPmRNA was decreased in EA group(P<0.01);compared with the model bound group,the expression of CGRPmRNA was decreased in EA group(P<0.05).(6)The level of5-HT:①Hippocampus:compared with the blank control group, the level of5-HT in hippocampus was ascended in model group and model bound group(P<0.05); compared with the model group,the level of5-HT in hippocampus was descended in EA group(P<0.05).compared with the model bound group,the level of5-HT in hippocampus was descended in EA group(P<0.05).②Colon:compared with the blank control group, the level of5-HT in colon tissue was descended in model group and model bound group(P<0.05); compared with the model group,the level of5-HT in colon tissue was ascended in EA group(P<0.05).③The ratio of5-HT:compared with the blank control group, the ratio of5-HT was ascended in model group and model bound group(P<0.05);compared with the model group,the ratio of5-HT was descended in EA group(P<0.05).Conclusion:1The method of neonatal maternal separation and acetic acid enema can prepare the stable IBS model animal.2Through the reaction of IBS visceral pain model rats sensitivity behavior and emotional psychological barriers study found that IBS animal model after preparation not only increased intestinal pain sensitivity, also cause their emotional behavior change.3Acupuncture can not only improve behavioral response, still can adjust intestines and the brain neurotransmitters and level changes accordingly4IBS model rats colon and associated with pain and emotional levels of neurotransmitters in the brain and change, is likely to be irritable bowel syndrome accompanied by emotional psychological barriers of one of the main mechanisms.
