Objective: Irritable bowel syndrome(irritable bowel syndrome,IBS)is a common chronic diseases without organs injury of the digestive system.Also known as berberine,berberine(Berberine,BBR)can be effective in the treatment of IBS,but the mechanism is not clear yet.Through the study,we will know the effects of Berberine on Visceral hypersensitivity and intestinal motility in Rats with Irritable Bowel Syndrome.We will investigate the possible mechanism of berberine for the treatment of IBS.Methods:(1)The model of rats with IBS could be established by using water avoidance stress(chronic water avoidance stress,WAS).Rats were divided into control group(Control group),water avoidance stress group(water avoidance stress group,WAS group),rifaximin group(Rifaximin group),low-dose berberine group(25mg/kg BBR group)and high-dose berberine group(100mg/kg BBR group).(2)The evaluation of the rat with IBS about the intestinal inflammation was as follows:we could observe the general state of the rats with IBS and HE staining were used to evaluate the pathological statement of terminal ileum tissue.(3)The evaluation of NF-?B signal transduction pathway: we could assess the NF-?B(P65)DNA binding activity and protein expression in the intestinal tissues by modified ELISA and Western bolt;we could assess the expression of mRNA gene about interleukin-1?(interleukin-1?,IL-1?)?interleukin-6(interleukin-1,IL-6)?interleukin-10(interleukin-10,IL-10)?transforming growth factor-?(transforming growth factor-?,TGF-?)?interferon ?(Interferon-?,IFN-?)?tumor necrosis factor-a,(tumor necrosis factor-a,TNF-a)by qRT-PCR.(4)The detection of Visceral hypersensitivity and intestinal motility: we could use the whole gastrointestinal transmission function and the intestinal transmission function to detect intestinal motility of the rats with IBS;we coulddetect the expression of BDNF and its receptor TrkB mRNA by qRT-PCR;we could detect the expression of TrkB protein by Western bolt.(5)The detection of interstitial cells of Cajal: we could detect the expression level of C-kit protein by Western bolt;we could detect the expression of C-kit mRNA by qRT-PCR.Results :(1)Water avoidance stress could make rats have visceral hypersensitivity,with the changes of the immune system and intestinal motility.All of these conformed to the characteristics of IBS.(2)Berberine could effectively improve the defecation of rats with IBS.Besides,the berberine could reduce the inflammation of distal ileum of rats with IBS.(3)Berberine could control the NF-?B signal transduction pathway that had aleady activated in the intestinal tract of rats with IBS.The berberine could reduce the expression of IL-1,IL-6,IFN-??TNF-a in the terminal ileum tissue,and promote the expression of anti-inflammatory cytokine IL-10,TGF-?.(4)Berberine could regulate the intestinal motility in rats with IBS effectively,with the inhibition of gene expression of BDNF and its receptors TrkB in terminal ileum tissues of rats with IBS.Berberine could inhibite the expression of TrkB protein.(5)Berberine could inhibit the expression of C-kit mRNA and C-kit protein in the ileal tissue.(6)Effect of the high dose(100mg/kg)of berberine on rats with IBS was significantly better than the low dose(25mg/kg)of berberine.Conclusion:(1)The water avoidance stress can successfully simulate the visceral hypersensitivity of IBS.(2)The activated NF-?B signal transduction pathway may be controled by berberine on the rats with IBS.Berberine can also reduce the expression of inflammatory cytokines and improve the expression of anti-inflammatory cytokines.To some extent,it can alleviate the inflammation reaction of intestinal tract on rats with IBS.(3)The expression of TrkB protein and BDNF and its receptorTrkB gene may be reduced because of the treament of berberine.Berberine can regulate the gastrointestinal motility and visceral sensitivity effectively.(4)The expression of C-kit protein and C-kit gene may be reduced because of the treament of berberine.Berberine can regulate the gastrointestinal motility and immune response effectively.(5)The effect of the high dose(100mg/kg)of berberine on rats with IBS is significantly better than the low dose(25mg/kg)of berberine.
