Objective To explore a new approach to clearing away human papillomavirus(HPV)infection, immune responses of system and genital tract were induced byintranasal immunization with HPV-E7and lymphotoxin(LT).Method Mice were randomly divided into four groups: PBS control group, LTgroup, HPV-E7group and HPV-E7+LT group. To determine the titers ofHPV-E7-specific antibody, serum and vaginal washing fluid of mice were collectedand assayed by enzyme-linked immunosorbent assay (ELISA). In vivo cytotoxicityassay,the clearances of target cells were measured by flow cytometry whichreflected the specific cellular immune responses of system and genital tract. Afterthree times of nasal immunization, draining lymph nodes of genital tract werecollected and the expressions of factors related to lymphocyte homing were detectedby immunohistochemistry.Results Specific antibodies were not detected in serum and vaginal washing fluid ofLT group. Titer of HPV-E7-specific serum IgG was1:1600in HPV-E7group and1:3200in HPV-E7+LT group. Differences were statistically significant. Titers ofHPV-E7-specific IgA were detected in vaginal washing fluids of HPV-E7group(1:20) and HPV-E7+LT group (1:40). And differences between them werestatistically significant (P<0.05). At the level of systemic cellular immunity, thekilling rates of LT group (6.16±0.97)%, HPV-E7group(26.25±5.04)%andHPV-E7+LT group (48.64±3.07)%were significantly different (P<0.05). At thelevel of genital tract, the killing rates of LT group(14.08±0.89)%, HPV-E7group(37.46±4.03)%and HPV-E7+LT group(69.12±9.01)%were alsosignificantly different (P<0.05). All factors related to lymphocyte homing inHPV-E7+LT group expressed (++). Compared with others, the differences werestatistically significant (P<0.05). Conclusions Specific humoral and cellular immune responses of system and genitaltract were induced by intranasal immunization with HPV-E7.All of these effects canbe effectively promoted by stimulating activation of lymphocytes and homing oflymphocytes through LT.
