Ramulus uncariae cumuncis is a kind of traditional commonly used Chinese crude drug.In research,scientists had found that alkaloids were known of one of the mian ingredients in it. Recently,there are increasing study on the alkaloids’s Uncariae effects on hypotentive effect.This paper studies on the extraction,content determination,hypotentive effect and inhibiting ACE effect. It is very important to realize the total alkaloid and physiological activities for further.The dissertation is consifituted by following parts:(1) Determination of the rhynchophylline contents by HPLC.(2) The orthogonal experimental design was used for the dominant selection and the content of rhynchophylline with HPLC determination as quantitative estimation standard.(3) Hypotentive effect of uncaria rhynchophylla on Spontaneous Hypertepsive Rats and inhibiting effect on ACE were performed.Results are the followings:(1) The HPLC system consisted of Hypersil ODS18(250 mm×4.6 mm,5μm)as the stationary phase and CH3OH:H2O=55:45(triethy-lamin 0.01moL/L,pH=7.5) as the mobile phase,the detection wavelength was 254nm.The results provide the rhynchophylline were on base line sepatated.The average recoveries and RSD were 98.35%,1.32%.(2) Reflux extraction is the best method of the three.And the quantity of ethanol,time of extraction and extraction times are the influence factors.The best process was set as follow:uncaria rhynchophylla was marinated 3 times with 90%alcohol and 4h at a time.(3) Tests with animal models,we find uncaria rhynchophylla with significant,lasting and slow effect on lowering blood pressure(P<0.05).But the inhibiting ACE effect is not significant.
