There are more than 500 species plants in genus of Polygala all over the world, but only 42 species and 8 varieties existed in China, mainly distributed in southwest and southern China. The root of Polygala tenuifolia or P. sibirica (Yuanzhi) is a commonly used traditional Chinese medicine (TCM), which acts as a sedative, expectorant and resuscitating agent, applied to the indication such as insomnia and dreaminess, amnesia and palpitation, staring spells, expectoration, pyccutaneous disease, and breast gall. Besides this, there are many other Polygala plants used widely in the folk. In this dissertation, the new progress of the sources, traditional usages, chemical constituents, pharmacological activities and metabolism of plants of genus Polygala has been summarized, which will supply a reference for the further research and development of this genus plants.The therapeutic basis matter of Yuanzhi, especially of the therapeutic matter in vivo has not been clarified clearly, so, in this dissertation, the vivo metabolism studies of total glycoside from "Yuanzhi"(TGYZ), the active fraction of Yuanzhi, as well as 3,6’-disinapoyl sucrose ester, one of the main active constituents of TGYZ, were carried on, and the preliminary metabolic process of them in vivo was illuminated. This would supply the reference for explaining the therapeutic basis matter of Yuanzhi and for the clinical usages.TGYZ was orally administrated at a dose of 500 mg/kg. The rats’urine and feces were collected separately, while the blood was taken from the eye socket. The sample pretreatment was operated according to the method described in the main text, and then HPLC-DAD-ESI-MS were carried on to analyze the metabolites in plasma and urine, and its metabolic pathway was also proposed. This study showed that the main metabolites were possibly generated from sucrose ester or onjisaponins conjuncted with cinnamoyl.The metablism of 3,6’-disinapoyl sucrose ester, the highest active constituent in Yuanzhi was studied in the vivo of rats. HPLC-DAD-ESI-MS method was established for the analysis of the metabolites in the plasma, urine and feces. The metabolic process in vivo was proposed. The reactions in vivo metabolic process includes hydrolysis of ester bond, reduce double bond of cinnamoyl and p-O-methylation of sinapoyl.In addition, the analytical method of TGYZ was established, and the structures of the main constituents were elucidated on the basis of UV and MS data obtained by HPLC-DAD-ESI-MS. The ultraviolet photometry and HPLC methods were used to analyze the stability of 3,6’-disinapoyl sucrose ester. The result showed that the stability of 3,6’-disinapoyl sucrose ester was influenced by the solvent type, temperature, pH and light. Among them, light is the most influenced factor. After disposition under the sunlight or UV lamp for a period time,3,6’-disinapoyl sucrose ester would transfer to be its cis-trans-isomers. So in the separation, purification and storage process, it is better to use alcoholic solvent, and to keep the sample in low temperature and dark place.
