| Objective:This study was set to illustrate the absorption,distribution,metabolism and elimination in rats of the main components from Acorus tatarinowii Schott and Polygala tenuifolia Willd,detected by ultra high performance liquid chromatography-tandem mass spectrometry(UHPLC-MS/MS).The essay explained the process of disposal of two Chinese medicines with similar pharmacological effects in rats,and confirmed the existence of material basis in corresponding target organs,which provided substantial basis for the pharmacological effects in the central nervous system(CNS)of Acorus tatarinowii Schott and Polygala tenuifolia Willd.Methods:Rats were orally administered with methanol extracts of Acorus tatarinowii Schott and Polygala tenuifolia Willd.The plasma,cerebrospinal fluid and organ samples were collected at different time points,the bile samples within 1 hour for a total of 12 hours,urine and feces samples collected every 24 hours for total 72 hours.All samples were analyzed by UHPLC-MS/MS after pretreated.Results:After oral administration of methanol extract of Acorus tatarinowii Schott in rats,the suspected compound cis-methyl isoeugenol(CMIE)was detected in biological samples with longer duration.Especially cis-methyl isoeugenol prototype was detected in the cerebrospinal fluid and brain tissue,it was speculated that it could pass by the blood brain barrier,play a role in the CNS.CMIE was also detected in feces and urine,but its metabolite not detected,it was not certain whether biotransformation occurred.After oral administration of methanol extract of Polygala tenuifolia Willd,suspected compounds 3,6’-disinapoly sucrose(DISS)and tenuifoliside A(TFS A)were detected in plasma,bile,liver,kidney,heart,and in cerebrospinal fluid and brain tissue,it was speculated that it can pass by the blood brain barrier arrived to the target organ.It provided the material basis for its pharmacological action in the CNS.Its absorption was rapid after the oral administration and stayed in the body for a long time,eventually excreted in feces and urine.The suspected phase I metabolite 3,4,5-trimethoxycinnamic acid of DISS and TFS A were detected in the liver,bile,feces samples after oral administration of methanol extract of Polygala tenuifolia Willd.It was speculated that the biotransformation of Polygala tenuifolia Willd took place in the liver.That means it can be absorbed into the blood system after the oral administration,metabolized by the liver and excreted by the bile into the intestinal,eventually excreted in feces.Absorption was quick after the oral administration extract of Acorus tatarinowii Schott and Polygala tenuifolia Willd,but had long action time in the body.The compounds CMIE,DISS,and TFS A were detected in various tissues and organs.The wide distribution confirmed there is a physical basis for pharmacological action.Conclusion:The dynamics of the methanol extract of Acorus tat arinowii Schott and Polygala tenuifolia Willd were systematically studied in rats through animal experiments combined with UHPLC-MS/MS analysis.The active components were detected in the systemic circulation and biological samples.The processes of absorption,distribution,metabolism,and excretion were described,to afford material basis for the pharmacological effects on the CNS of two Chinese medicines. |
