Purpose: The purpose of this study was to clarify the effect of endurance exercise oncaveolin-3content of cellular membrane which was treated by CsA. In the concrete, we usedbiochemical index and the changes of ion concentration in the extracellular to discuss theeffect of caveolin-3on exercise-induced cell protein metabolism, cell signaling pathways inendurance exercise, mechanisms of changes of caveolin-3, and to provide a theoretical basisfor endurance exercise-induced changes of the cell structure and the recovery of fatigue.Methods:6weeks male Sprague–Dawley rats were used in this study. They were randomlydivided into four groups, control group (C), training group (T), Cyclosporine group (S) andCyclosporine-training group (ST). There were ten rats in each group. After two weeks ofadaptive exercise, rats were trained at a speed of28m/min,10%grade on the treadmill for13weeks. The training was6days in each week, and each daily session consisted of running for1h. From10weeks of training rats of Cyclosporine group and Cyclosporine-training groupwere injected CsA. After full training protocol, rats were killed for harvesting aortic blood,gastrocnemius, soleus, extensor digitorum longu. Then gastrocnemius, soleus, extensordigitorum longu were made liquid through homogenate and centrifugal. To measurecaveolin-3content of cellular membrane, the concentration of Ca2+, CK and LDH in the blood,LDH in the organization, NO and NOS, we used immunohistochemical method, fluorescentindicator method, rate method, and nitrate reductase method.Results: After endurance exercise, there was no difference of concentration of Ca2+in theintracellular between control group (C) and Cyclosporine group (S)(P>0.05). Theconcentration of Ca2+in the training group (T) was significantly higher than in the controlgroup (C)(P<0.01), and the concentration of Ca2+in the Cyclosporine-training group (ST)was higher than control group (C)(P<0.05).NO in the Cyclosporine group (S) was higher than control group (C)(P<0.05).There wasno difference of NO between training group (T) and control group (C), but NO of the traininggroup (T) has an increase trend. NO in the Cyclosporine-training group (ST) wassignificantly higher than control group (C) and training group (T).Compared with control group (C), NOS in the Cyclosporine group (S) and the traininggroup (T) is higher. There was no difference of NOS between Cyclosporine-training group(ST) and control group (C)(P>0.05).CK was not influenced by exercise and CsA. Compared with control group (C), LDH in the blood of Cyclosporine group (S) is higher(P<0.05).Caveolin-3content in the extensor digitorum longu of the order from highest to lowestwas C, ST, T, S. Caveolin-3content in the soleus of the order from highest to lowest was C, T,S, ST.Conclusion:(1) CsA was cause skeletal muscle cell membrane injury.(2) Concentration of the intracellular Ca2+rose after long time endurance exercise.Separate CsA intervention on cytosolic Ca2+concentration changes had little effect. Endurance exercise and CsA interaction, intracellular Ca2+concentration rose.Effects ofcytosolic Ca2+concentration was higher by endurance exercise than by CsA.(3) Both NOS and NO were rise in skeletal muscle tissue under the effect of CsA.NOS in muscle tissue rose after long time endurance exercise,NO reduced becauseof consumption in substrate synthesis.Under the situation of both CsA and enduranceexercise,NOS had unobvious change and NO increase to protection of cell membrane.(4) LDH increase under the effect of CsA,CK restored to its originai level.LDH recovered more slowly than CK,more suitable to reflect damage.(5) Caveolin-3reduced in different kinds of muscle fiber types,Structural proteingot degradation, Caveolin-3mediated signal pathway was interferenced under effect ofboth CsA and long time endurance exercise.The effect was more significant in slowmuscle than in fast muscle from CsA.
