Dehydrin is one of late embryogenesis abundant proteins, the molecular weight isbetween9and20kD. It can be induced greatly under environmental stresses, such as cold,drought and salt stress. It is a kind of hydrophilic protein which with the characteristic ofthermal-stability.Dehydrin could reduce the drought injury on plant by combining withmembrane lipid. It acts as a molecular chaperone when the plants suffer the water stress.In my study, we did some analysis with bioinformatics software firstly. We found therewere a great number of cis-acting elements which were associated with adversity, such asTGACG-motif(involved in the MeJA-responsiveness), MBS(involved in drought-inducibility),LTR(involved in low-temperature responsiveness), ABRE (involved in the abscisic acidresponsiveness), SARE(involved in salicylic acid responsiveness), GARE-motif (gibberellinresponsive element). According to the distribution of the cis-acting elements on the promoter,we got seven5’ flanking deletion promoters,they were called sp5(736bp), sp6(541bp),sp7(431bp), sp8(395bp), sp9(257bp), sp10(184bp), sp11(164bp).We made use of thesedeletion fragments connecting with vector pBI121which carrying a reporter gene gus. Thenwe transferred them into the wheat callus with the method of microprojectile bombardmentfor transient expression assays. on the basis of histochemical analysis and fluorescentquantitative analysis of GUS staining, we predict the promoter wzy1-2is an induciblepromoter,wich showing the characteristic that induced by ABA preferentially.At the sametime, when under the inducing of salicylic acid and gibberellin, the expression was alsoenormously. It was just a small amount of expression under the methyl jasmonate and droughtstress-induced.What was more, it acted as nearly no expression under the inducing of lowtemperature.
