Echinacea (Echinacea purpurea) belongs to Echinacea genus of composite family, which is by far the world has found a very small number of immune enhancement and anti-Inflammatory herbal dual role, one has wide range of treatment and stable results. In recent years Echinacea purpurea was introduced in some regions of China, its chemical composition and pharmacological activities have been studied deeply. With the appearing of the health care products and preparations of the introduced Echinacea purpurea, people attach more importance to the study of their quality. Nowadays, the quliaty control of Echinacea purpurea and their preparations mostly depends on the standard of caffeic acid derivatives, which can characterize the quality of Echinacea purpurea.So we systematically studied on the caffeic acid derivatives from the Echinacea purpurea.we used thin-layer chromatography(TLC), HPLC-DAD, to track and locate which part were the subtances in. and found that Caffeic acid derivatives mainly exist in the n-butanol phase.Seven compounds were isolated and purified by (TLC), SephadexLH-20, column chromatography on ODS,(HPLC) and semi-HPLCthose compounds, by MS,’H-NMR,13C-NMR and other spectroscopic methods combined with the physical and chemical properties identified as chlorogenic acid (1), ferulic acid (2), caffeic acid (3), chicory acid (4), Caffeoyl-feruloyl-(2R,3R)-(+)-tartaric(5), uracil (6), thymidine (7). These compounds are known compounds.The extraction and separation process, simple operation, low production costs, pollution and the use of Jess toxic organic solvents, easily recycled. Preparation of high purity, health food and herbal medicine can be used for the analysis of standards.Meaning of this experiment:Through Systematic isolation and identification of caffeic acid derivatives, specifically in the composition of such components, lay a foundation for the future the research of the quality control. Preliminary exploration of the caffeic acid derivatives of the extraction and separation process, the for the future of the foundation.
