Echinacea purpurea, which belongs to Echinacea genus of Composite family, is native to the central and western regions of the North America. Echinacea involves nine species and two variants, and Echinacea purpurea, Echinacea palliday,Echinacea angustifolia have been developed for drugs.As a immunostimulant and immunoregu-lant, Echinacea purpurea has anti-inflammatory, antibacterical, antiviral, antitumor antioxidant and insecticidal effects.In1970s, after Echinacea purpurea was introduced in some regions of China, its chemical composition and pharmacological activities have been studied deeply. With the appearing of the health care products and preparations of the introduced Echinacea purpurea, people attach more importance to the study of their quality. Nowadays, the quliaty control of Echinacea purpurea and their preparations mostly depends on the standard substance of Cichoric Acid, which tends to be oxidized and is hard to be isolated and purified. Therefore it’s important to search for another standard substance. Previous work showed that, as an essential component of the Echinacea purpurea alkamides have functions of anti-inflammatory and antioxidant. It’s reported that alkamides were used as the standard substance during the quality control of the Echinacea purpurea in America. So we study on the alkamides of Echinacea purpurea and try to prepare the standard substance of the alkamides, expecting to provide a reference to the quality control of Echinacea purpurea.We systematically studied on the alkamides from the Echinacea purpurea collected from Tongcheng of Anhui province. By thin-layer chromatography, HPLC and Diode Array Detector, we tracked and located which part were alkamides in. Ten compounds were isolated and purified by thin-layer chromatography(TLC), SephadexLH-20, column chromatography on silica gel(CC),high performance liquid chromatography(HPLC) and and semi-HPLC. On the basis of entensive spectroscopic analysis and comparison their spectral data with these reported in literatures, their structures were identified as deca-2Z,4E-diene-8,10-diynoic acid isobutylamide(1), undeca-2.E,4E-diene-8,10-diynoic-acid-2-methylbutylamide(2),Trideca-2E,7Z-diene-10,12-diynoic acid iso-butylamide(3), dodeca-2E,4E,8Z,10E-tetraenoic acid isobutyla-mide (4)and dodeca-2E,4E,8Z,1OZ-tetraenoic acid isobutylamide(5), undeca-2E,4E-diene-8,10-diynoic acid isobutylamide(6), undeca-2E,4Z-diene-8,10-diynoic acid isobutylamide(7),dodeca-2E,4E-diene-8,10-diynoic acid isobutylamide(8) and dodeca-2E,4Z-diene-8,10-diynoic acid isobutylamide(9). Copound2was obtained from the plant for the first time.
