Lotus leaf rescource is very rich in China, Jiangxi Guangchang is known as the landof White Lotus which was first introduced in space breeding. However, there have beenfew reports of the study for Space Lotus. with the theme of green and healthy, nuciferineand procyanidins, two active ingredients from Space Lotus were choosed as the objects ofthe research,and we opitimized the process that Space Lotus used as tea.The content of nuciferine had significant differences at different growth stages anddifferent harvest time,and it was highest at early august. But even in this period,thecontent is not the same in varying laonen degrees,in newborn leaves has the maximumcontent0.55%.After pretreatment,the lotus leaves were dealed with different dryingtemperature to compare the loss of nuciferine,it was found that the minnimun loss wasobtained at50℃blast dried with hige drying efficiency. To further verify the stability ofnuciferine,we considered the effects of heating and light on nuciferine,and the resultsshowed that it can be much more stable in low temperature and dark place. We can mincethe leaves and make it into bag-soak-tea. leaves laonen, soak time, extraction times anddifferent types of acids all of them have influenced the active diffusion of nuciferine.whenchoosed new leaves, add0.1g of citric acid,1g lotus leaf powder a pack with100mlboiling water and soak for3times, each time20min, the nuciferine will completely soakout with a content of0.6%.The process of nuciferine extraction and separation from Space lotus: addingphosphoric acid until pH to2then heating extract several times. The optimal extractionconditions were found as follows:20:1material-to-liquid ratio,50℃extractiontemperature,1.5h extraction time and three extractions. under this optimal conditionnuciferine content from space lotus leaves was0.83%. HPD-100type of macroreticularadsorbent resin was used to enrich the nuciferine, and nuciferine crude content can reachmore than9%. Further, precipitate nuciferine with alkaline we got a pale yellow nuciferinewith a purity of20.7%, and then extracted with ethyl acetate final nuciferine contentincreased to50.14%.The process of procyanidins extraction and purification from space lotus leaves:Thesingle factor experiment and response surface method was applied to optimize theextraction of procyanidins from space lotus leaves. The optimal extraction conditions werefound as follows:55%ethanol as extraction solvent,80℃extraction temperature,pH3.0,20:1material-to-liquid ratio,1.5h extraction time and twice extraction. Resultsindicated that procyanidins extraction from space lotus leaves was78.65mg/g under thisoptimal condition. HPD-400type of macroreticular adsorbent resin was used to adsorb anddesorpt the extract. The maximum adsorption volume was3.5BV, with a adsorption flow rate of1.5ml/min, and the parsing was washed by50%ethanol with a analytic flow rate of1ml/min.. We condensed the eluent by rotary evaporation and dried the rest solution1h in avacuum at40℃to get the brown-red powder. The96.21%purity of proanthocyanidinswas detected by Vanillin-sulfuric acid and spectrophotometer method.
