In recent years, people have paid more and more attention to food security. Food-borne pathogens are major factors of food safety incidents. Food-borne pathogenic bacteria are one of the important parts in food testing. With the development of technology and the requirements of food testing, the testing technology is constantly improved in the direction of accuracy, efficiency, and diversification.Nano-technology is a new study in the world. And it is widely used in the biological area, including biochemistry, cell biology, molecular biology, genomics, proteomics, medical diagnostics, drug screening and so on.The experiments are based on National food safety standard Food microbiological examination:Salmonella (GB/T4789.4-2010) and local standards (DB31/T455-2009). We have constructed a detection kit used to detect Salmonella and studied production technology, quality control standard, and verification method. The Salmonella detection kit consist of four parts:immune magnetic beads coated with silica and modified with Rabbit anti-Salmonella polyclonal antibody, immune quantum dots modified with Rabbit anti-Salmonella polyclonal antibody and Buffer1(pH=6.0) and Buffer2(pH=7.2). In this study, we determined the information of quality control of the kit, the optimal size of immune magnetic beads, the best optimum concentration of liquid adding amount and frequency of use.Ther detection limit of Salmonella was103CFU/mL. The kit could be stable and reliable for three months when stored at4℃. The experimental results show that the kit can specifically detect Salmonella, and realize the visualization of Salmonella detection.In addition, this study designed a biological sample magnetic enrichment equipment BN10-CXY-1, which was able to shorten the detection time greatly, purify rapidly the concentration of Salmonella in large volume sample. When used with the IMS-QDs Kit and BN10-CXY-1, we could realize the automatic detection of biological samples, improve the sensitivity, and meet the needs of rapid detection of food safety field.
