It was firstly ranked in source directory of foodand medicine by The Ministry of Health. It has a lot of benefits such as moistening lung tocough, regulating spleen and stomach, enhancing the immune.etc. During the productingprocess of lily starch, the residue is often abandoned, causing great waste of resources. In thispaper, total saponins were selected as research object after the preselection of activeingredient in lily. For a good result, the optimal conditions of the extraction and purificationprocess of lily total saponins was investigated. And the influence on immune function of themice was preliminary discussed. The main content can be drawn as follows:1) The ultrasonic-microwave synergistic extraction technology of total saponins wasinvestigated with total saponins yield. Effects of ethanol concentration, liquid-solid ratio,microwave power and extraction time on total saponins yield were determined. Based on theresults of single factor tests, as well as Box-Behnken experiment principle, the four factorsand three levels response surface method test was designed and total saponins yield wasdetermined as response value. The optimum extracting parameters was obtained as follows:ethanol concentration(v/v) of77%, liquid to solid ratio of11mL/g, microwave power of503W, and extraction time139s. Under these conditions, the total saponins yieldand extraction rate were6.36‰and87.4%, respectively.2) Macroporous resin was used to purify total saponins. Three types of macroporousresins,namely S-8, AB-8and X-5were compared by their aspects of the physical properties,static adsorption and desorption rate and static adsorption kinetics curves. Macroporous resinAB-8, which have the highest value of static adsorption, the faster rate to attain the staticadsorption equilibrium, and the higher elution rate, was selected to purify the total saponinscrude extracts solutions. The technology conditions of purifying total saponins bymacroporous resin AB-8were investigated through the pH and salt ion concentration of staticadsorption, the concentration and flow velocity of the sample solution, the volume and flowveloscity of eluant. The optimum conditions for purifying total saponins were as follows: theconcentration of sample solution2.0mg/mL, pH7.0, salt ion concentration2.0mg/mL, flowveloscity3BV/h. The elution conditions were eluant70%ethanol, flow veloscity2BV/h.Under these conditions, the purity of the total saponins was enhanced from35.2%to64.3%.3) The influence of the preliminary purified total saponins on the immune function ofmice was discussed from the following four aspects: the thymus index and spleen index,lymphocyte proliferation, antibody cell generation and serum hemolysin. The experimentalresults were as follow: spleen index of mid-dose and high-dose groups and thymus index ofmid-dose group were significantly difference compared with control group. There weresignificantly differences between mid-dose and high-dose groups and control group inlymphocyte proliferation and antibody cell generation experiment.
