Citrus limon is the most frequently used fruit in food industry, and it has been a focalpoint for healthcare products research. Functionally, lemon has hypoglycemic,antibacterial,antioxidant and other bioactivities. In this study, we establish methods fordetermining the Total saponins, Limonin compounds, The trace element zinc, sod activity,in lemon,and functionally peptide in lemon was investigated in this paper.In order to determine the total saponins in lemon, the method for separation andpurification of total saponins from lemon by macroporous adsorption resin and determinationfor total saponins by UV spectrophotometry are researched. The total saponins were coloredby solution containing vanillin and perchloric acid and then detected by UVspectrophotometry in560nm. Ginsenoside Re was taken as control article.The total saponinsin10-flod dilution of lemon sample is27.1mg/L.The total saponins in20-flod dilution oflemon sample is27mg/L. In the40μg~μg scope, it has a good linear relationship (R~2=0.9997).The average recovery rate are102%,108.5%and111%, RSD=2.38%,1.22%. Inorder to determine the sod in lemon, the method for separation of SOD from lemon by80%saturation ammonium sμLfate initially and determination for SOD by Marklund areresearched.The activity of lemon SOD sample is25.99u/mL.The standard solution of SOD is332.42u/mL.RSD=2.15%,2.16%.In order to determine the limonin in lemon, the method for separation of limonin fromlemon by SFE and determination for limonin by HPLC are researched. In the40mg/L~80mg/L scope, it has a good linear relationship (R~2=0.9993). The limonin in lemon sampleis61.03mg/L.The stability of lemon sample is20h.RSD=1.04%,1.63%.In order to determine Zn in lemon, the method for determination for Zn by microwavedigestion and atomic absorption spectrometry are researched.The Zn in lemon juice is1.13μg/g. In the0.05μg/mL~0.6μg/mL scope, it has a good linear relationship (R~2=0.9993).The average recovery rate are91%,102.25%and105%.The Relative uncertainty ofdetermination of sample repeatability and the Zn content of the standard solution and the leastsquares fit the standard curve content are0.59%,0.5%,0.274%. The standard solution of Zn inthe preparation of pipette and volumetric flask and sample digestion solution constant volumehas smaller impact on the determination.The impact of weigh is neglible.In order to research peptide in lemon, the method for determination for separation andpurification of peptide from lemon by Khanan and HPLC are researched.Sample withninhydrin solution reaction,the Result is colorless to purple to blue.By RP-HPLC elutinglemon sample and recombinant human insμLin,the Results show that the absorption peaksapprar at almost the same retention time.The selection for gradient elution conditions of lemon samples,the Results show that the better gradient elution conditions:mobile phase flowrate of0.8mL/min,0-1min:A solution:90%, B solution:10%,1-30min: A solution:40%, Bsolution:60%...
