| Condions on enzymatic extraction of biological POT saponins and purification to enzymolysis products of POT saponins were studied.Through the experiment,the optimal conditions for the enzyme cultivation were explored in:beginning with Absida sp.O8s,enzymatic extraction of biological POT saponins the optimum:5g POT powder used 40mL 0.02mol/L phosphate buffer at pH 6.0;soak 4 hours at a temperature of 50℃ conditions with 40mL coarse enzyme,pH6.0 enzyme reaction after 12 hours at room temperature to three times the volume of 95%ethanol,at 40℃ or 45℃static eight hours,filtration,the filtrate evaporated to no alcohol flavor.Residual liquid distilled water to the volume to 100mL.The purification of the products by AB—8 macroporous resin were studied in this paper.lt was showed that the adsorption capacity of AB—8 resin column was 53.16 mg/mL and the eluting reagent was solution of 80%ethyl alcohol.Finally the eluted juice was condensed and desiccated.The yield rate was 85.49%.After purified by AB—8 Macroporous Adsorptive Resins,total POT Saponins was ed for the second time through Gel Silica Colume.Then the detection of POT Saponins through HPLC showed the result:according to the different volume-ratio of Trichlormethane and Methanol,the POT Saponins was abstracted in seven constituents.When the volume-ratio reached 9.7:0.3,the constituent marked E was abstracted.The E was detection through HPLC.The result showed that preparation of POT Saponin marked C was contained in E,and the purity quotient is 70.8%. |
