As we know the concept of organic synthesis that the organic reaction can only be carried out in organic solvent. However, because organic solvents are harmful to the environment and human, Now many novel and ecofriendly solvents such as fluorous media, scCO2, Ionic liquid and water have attracted the attention of organic reactions. As the water non-toxic, renewable, low-cost, and so more and more used in organic synthesis. This paper focuses on research in the aqueous synthesis of pyrimidine and quinazoline. this thesis can be divided into three parts.Green chemistry, also known as environmentally benign chemistry name suggests biological and environment-friendly, harmLess and sustainable development, through the mechanism of chemical reactions to design a chemical reaction to reduce or eliminate the using of toxic and hazardous substances. Green chemistry require the reaction to comply with:atom economy, efficient, rapid and mild reaction, the solvent should be ecofriendly.In Part two, We have developed a one-pot, metal-free-water media reaction that provides of metal-free catalytic catalytic efficient one-pot synthesis of4-amino-2-(Alkylthio)-6-phenyl-5-Pyrimidinecarbonitrile derivative directly from aqueous Thiourea, halogenated, aromatic aldehydes and malononitrile. The method is technically straightforward, robust, versatile, and high yielding. Using a combinatorial approach, we have prepared an array of4-amino-2-(Alkylthio)-6-phenyl-5-Pyrimidinecarbonitrile derivative.In part three, We have discovered a novel and efficient CUSO4.5H2O catalyzed1,3-dipolar cycloaddition of quinazolin-3-ium-3-yl(tosyl)amide with Alkyne which provide a mild,highly regioselective method. We can conveniently obtain2-phenylpyrazolo[1,5-c]quinazoline derivative.
