Isodon excisa (Maxim) Hara. belongs to family Lamiaceae. Isodon excisa has been used as folk medicine for the treatment of respiratory and gastrointestinal bacterial infections, sore throats, inflammation, and cancer in China. Researchers at home and abroad study on the chemical composition of the Isodon excisa. Several diterpenoids、triterpenes、flavonoids and other compounds were separated and identified, and focus on against tumour, antibacterial, antiviral and so on.This paper extract and isolate and active studies on the dry leaves of Isodon excia of the Changbai Mountain. It has been less reported the tyrosinase inhibitory activity and free radical scavenging activity of Isodon excisa in the literature, in this experiment, the Tyrosinase inhibitory activity and DPPH radical scavenging activity are used as a guide. The silica gel column chromatography ODS reversed phase column chromatography, PTLC, and semi-preparative HPLC and other separation methods were used to isolate the ethyl acetate extract of the Isodon excisa (Maxim) Hara., and12compounds were isolated. Modern spectral analysis techniques (NMR、 MS、IR and so on) was used to establish the structures of four compounds, Wangzaozin A, Glaucocalyxin A,2α-hydroxyursolic, maslinic acid.In this paper, all the different fraction of the extracts and components were tested by the tyrosinase inhibitory activity and DPPH radical scavenging activity experiment. The result of test showed that the extracts and of fractions inhibited the DPPH radical and tyrosinase, Some of the components of DPPH inhibition rate associated with compositional polarity are presented, namely with the increase of the polarity, the inhibition rate increased as well. The inhibition ratios of7fractions against DPPH radical were100%such as IE1.5.7.9、IE1.7.2and other13fractions were over97%.while8fraction against tyrosinase was90%and the inhibition ratios of fraction IE2.12.6.4was100%best. IE2, IE3and other18fractions have two kinds of strong activity. The inhibition ratios of fractions were over80%. The compounds were tested on the DPPH radical scavenging activity、tyrosinase inhibitory activity、 dual-Lueiferase reporter gene and MTT experiment. Our work is to develop new drugs to provide the basis of experimental data.
