Thiol compounds widely exist in the organism, antioxidant, anti-aging, detoxification of heavy metals, scavenging free radicals and other reactive function. Thiol compounds can make the thiol oxidation during the process of extract extracellular environment, lead to the determination results of severe low thiol compounds, even missed detection. In recent years, the extraction and determination of thiol compounds in biological samples must be approved by, the cell wall-breaking, extraction, derivatization and to protein before processing. In this study three different pretreatment methods for the determination of thiol compounds are discussed in this paper, the cell wall-breaking and derivative into an organic whole of synchronous derivatization method for extraction of the sample extract has the highest sensitivity. Using cell wall-breaking and derivative synchronous extraction method to get accurate information about the content of thiol compounds.Chapter1:PrefaceIntroduces the research situation and thiol compounds of thiol compounds analysis methods, the topic of this paper and the main researching contents of this subject are proposed。Chapter2:Detection of Thiol Compounds in the Earthworm using DTNB as Derivatizing ReagentUsing derivatization reagent DTNB, using UV-Vis spectrophotometric method for determination of total sulphur content in the earthworm and selective HPLC method determination of GSH, Cys. GSH, Cys and total sulphur content were0.6268μmol/g,0.8511μmol/g and4.5087μmol/g. According to the experimental data, the earthworm contains relatively high content of Cys, that earthworms have high ability of assimilation metabolism of sulfur in the soil and make it into Cys, for GSH biosynthesis and MTs provides abundant source of amino acids containing sulphur.Chapter3:Determination of total Thiol Compounds and GSH in Earthworm using Spectroscopic MethodThis study used4-DPS and OPA derivatization reagent, spectrometry total sulphur and GSH content in earthworms. From the experimental results, the earthworm, rich in GSH and sulphur compounds in earthworm and thiol compounds of heavy metals in the body have the strong coordination role, the earthworms are used for bioremediation of heavy metals pollution provides important chemical basis.Chapter4:Detection of Thiol Compounds and GSH in the Yam using Derivatizing ReagentUsing4-DPS and OPA derivatization reagen twith three different pretreatment methods, respectively for UV-Vis and fluorescence spectrometry determination of total sulphur and GSH in the yam. Rich thiol compounds found in yam, synchronous derivative spectrometry total sulphur and GSH content of0.4343~0.4556μmol/g and0.1649~0.1711μmol/g. Method of test line, and recovery is to achieve the requirement of the sensitive and accurate measuring.
