Capillary electrochromatography monolithic column is that functional monomer(Allyla-mine-β-cyclodextrin and glycidyl methacrylate acting), crosslinking agent and initiator arebonded on the inner surface of capillary. There is a network structure with large and uniformpore size in the monolithic column. The advantages of capillary monolithic column are easypreparation, low column pressure, high efficiency, excellent permeability, high rapeatabilityand rapid separation. Capillary monolithic column is widely used in Capillaryelectrochromatography.In this paper, reactant ratios,the reaction time and temperature were studied andoptimized. Under the optimized preparation conditions, the capillary chiral monolithic columnwas prepared. The prepared column has better permeability and better separational capability.In capillary electrochromatography (CEC) mode, The as-prepared monolithic column wasevaluated. The influences of buffer pH value, temperature and voltage on separation wereinvestigated. At the same time, reproducibility and stability were studied by separatedDL-Phenylalanine for seven times. The results show that the RSD of L-Phenylalanine was1.53%and the RSD of D-Phenylalanine was2.39%.The RSD of separation was2.92%.Theoretical plate number of the column was more than85488plates/m with thiourea aselectricity seepage markers. The monolithic column was used for continuous16h andintermittent3months with keeping good column efficiency. Under the optimal CECconditions, Norfenefrine hydrochloride, Benserazide hydrochloride, Cephradine, Guaifenesinreached baseline separation.
