With the rapid development of Raman spectroscopy and Surface-enhanced Raman spectroscopy (SERS), those technologies have been used in the biomedical research. In this thesis, the new method of semen quality analysis base on the Raman spectroscopy and SERS technology to research the human seminal plasma in preasented, which to avoid the current clinical shortage. The main research results as follows:1. Using Raman spectroscopy to characterize the seminal plasma specific spectral features and PCA-LDA analysis methods can be used to detect and differentiate semen samples by rapid and simple examination of the corresponding seminal plasma obtained from the normal and abnormal subjects. The diagnostic sensitivity and specificity was73%and82%, respectively.2. A polarized SERS method was firstly developed for seminal plasma biochemical analysis. The results from this exploratory study showed, with the non-polarized laser excitation, both the sensitivity and specificity were around70%, whereas high diagnostic performance was achieved by left-handed circularly polarized laser excitation (sensitivity of95.8%, specificity of64.9%).3. Citric acid detection in clinical seminal plasma ultrafiltrate samples was performed, and the intensity of Raman specific peak demonstrates a good linear correlation with citric acid concentrations determined by the enzymatic method. Our results showed that Raman spectroscopy has the potential of being applied to detect concentrations of citric acid in seminal plasma in clinic.Our work indicated that, the use of Raman spectroscopy and SERS in seminal plasma ananlysis has advantages of convenient and feasible, etc. The spectrums of the seminal plasma of the normal and abnormal were distinguished well when combined with the analysis method of PCA-LDA and the technology of laser polarization. The results showed that the Raman spectroscopy and SERS might provide a new kind of method for analysis the quality of semen.
