Resveratrol(RES) is a naturally occurring polyphenol present in grapes, peanuts and polygonum cuspidatum. The RES has been proved to have the function of anti-tumour, anti-inflammation, immune regulation, anti-oxidation and preventing anti-cardiovascular disease. In this article, we study into the anti-aging and anti-oxidation funtion of RES on C.elegans and mice, and its mechanisms.Firstly, we study the anti-aging effect of RES on C.elegans and its mechanisms at molecular level.50mg/L and100mg/L RES have been used to test its effect on elegans’ life-span under the temperature of25℃and35℃. This experiment aims at studying the molecular mechanism of the RES extending C. elegans’ life-span through regulating the expression of daf-16, sod-3and hsp-16.2genes, which provides strong evidence for anti-aging function of RES. The results show that the RES of100mg/L can obviously extend the life-span of C.elegans at normal and heat pressure state (p<0.05). Gene expression analysis shows that these three genes are all up-regulated, which may be involved in the anti-aging function of RES.Secondly, hyperlipidemia mouse model is established to explore the anti-oxidation of lipid in vivo and the genes expression involved.3kinds of concentration of res have been set, including60,30and10mg/Kg/day. The Zhibituo is used as a contrast in modulating the serum lipids in hyperlipidemia mouse. The TC, TG, HDL-C and LDL-C concentration in serum are tested to evaluate if the model is established successfully. The SOD and MDA have been tested to evaluate the anti-oxidation ability of RES. Relevant genes including ACC1,CYP7A1, CPT-1and sod-3are studied using RT-PCR method, We conclude from the results that RES can decrease the concentration of TC,TG and LDL-C in serum, the effect caused by high-dose RES is most obvious(p<0.05). Gene expression analysis shows that the ACC1and CYP7A1are up-regulated while the CPT-1is down-regulated, which may contribute to explain the phenomenon above; High-dose RES can highly increase the SOD and decrease the MDA in liver of mice (p<0.01). It can be concluded that the RES has the function of anti-oxidation of lipid. Gene sod-3expression analysis also contributes to it.
