In this paper, fresh wolfberry as raw materials,using single factor test,orthogonal test and Box-Behnken experimental design,Optimization the process of organic solvent extraction,magnetic-assisted organic solvent extraction,ultrasonic and microwave-assisted organic solvent extraction; wolfberry Pomace Carotenoids which was absorbed and desorption by macroporous resin,and researching the stability of wolfberry pigment.The main research studies were as follows:1. The results of the organic solvent extraction showed that:orthogonal test depanding on the single factor results was measured to determine the optimal parameters of the organic solvent extract of wolfberry carotenoids:mixed solvent (ethyl acetate and ethanol) was best organic solvent,temperature of55℃, the extraction time was4h, particle size of80mesh, the liquid ratio was1:25,under this conditions,the content of carotenoids was141.56mg/100g.2. The magnetic-assisted organic solvent extraction processing was optimized trough single factor and Box-Behnken response surface analysis. the extraction process parameters as followed:magnetic stirring time19min, magnetic stirring speed800r/min magnetic stirring temperature41℃, the content of carotenoids was144.21mg/100g, and the relative error with the theoretical value146.18mg/100g was1.34%。3. The ultrasonic and microwave-assisted organic solvent extraction processing was optimized trough single factor and Box-Behnken response surface analysis. the extraction process parameters as followed:ultrasonic output power282W, the particle size80mesh, microwave power275W, the content of carotenoids was134.84mg/100g, and the relative error with the theoretical value (136.98mg/100g) was1.56%4. To refine carotenoids from wolfberry pomace by macroporous resins, the static adsorption and desorption performances were investigated among HP20、S-8、AB-8、D101and X-5resin. The experimental result showed that X-5resin under the condition of adsorption time120min,pH4,35℃has the highest adsorption rate was421ug/g, using Ethyl acetate and the desorption rate was81.7%; Freundlich and Langmuir equations were used to model the adsorption isotherms at equilibrium in the range of25-45℃,the result show that the both model fit the experimental date. The adsorption process of X-5resin is a monomolecular adsorption, but not belong Shipping adsorption;X-5resin dynamic adsorption and desorption test results show that:When fixed Sample liquid flow rate between1.5-4.5ml/min, Macroporous resin can reach8times bed volumes (160ml), Under similar conditions the dosage of Eluent as7.5times bed volumes (150ml). 5. The stability of the pigment showed that:High temperature,light,oxidants, Fe2+,Fe3+, Al3+and Cu2+had a larger role in the degradation and destruction of Carotenoids; reductant and consumption raw had no significant impact on the stability of carotenoids; Antioxidants can slow down the degradation of the pigment.In conclusion, the content of carotenoids in wolfberry pomace is more abandent and palyed an improtent role in protected human health,Researching the extraction of carotenoids from wolfberry pomace not only extend the production chain,reduced the environmental pollution,but also had a great significance to promote the healthy and sustainable development of wolfberry industry.
