In order to overcome the defects of the existing biological glue,which is the deficiency ofdrug-load, this research adopt α-cyanoacrylate as the pharmaceutical carrier to prepare5-fluorouracil drug-loaded microspheres, and penetrate the microspheres into theα-cyanoacrylate monomers to form the medical adhesion of drug-loaded. The investigationand study on the characterization of5-Fluorouracil/α-cyanoacrylate drug-loaded microspheresas well as drug-loaded medical adhesive is included. The main work and results in the thesisare listed as follows:(1) Preparation and discussion of5-fluorouracil/α-cyanoacrylate drug-loadedmicrospheres.Electrostatic spray method is adopt in the preparation of5-fluorouracil/α-cyanoacrylatemicrospheres. Compared with several different processes of preparing the microspheres, abrief discussion about the impact electrode voltage, the distance between electrodes, flowspeed of the solution to preparing microspheres are introduced. The selected voltage is20KV,the selected distance between electrodes is10cm, the speed of the solution is3ml/h to preparemicrospheres that the mass ratio of5-fluorouracil to α-cyanoacrylates is1/2, the surface ofmicrosphere was observed by scanning electronic microscope (SEM), the grain size and sizedistribution was tested via the Malvern Lazer Particle analyzer, the structure was studied byinfrared spectrometer and differential scanning calorimetry(DSC). The results showed thatdispersity of the microsphere is very good, the surface was pleated, distribution ofmicrospheres’ size were uniform. The shape is similar to flower, with the average diameter ofmicrospheres--500nm. In the microspheres5-fluorouracil dispersed uniformly, with nophenomenon of gathering or crystallization which indicates5-fluorouracil has highcompatibility with α-cyanoacrylates. Test about storage stability of microspheres showed that:The microspheres is more suitable to be stored at low temperatures.(2)Preparation and Characterization of drug-loaded medical adhesiveThe drug-loaded microspheres dispersed uniformly into α-cyanoacrylate under theprotection of nitrogen, which has drug-loaded medical adhesive. The materials is tested inaspects of tensile strength, impact strength and bending strength of drug-loaded medical adhesive. Interaction between microspheres and medical adhesive was characterized viainfrared spectrometer and differential Scanning Calorimetry (DSC). Finally with ultravioletspectrophotometer the drug release behavior of the drug-loaded medical adhesive wasinvestigated and studied. The result showed that: medical adhesive was reinforced under thepenetration of microspheres. In a certain aspects, mechanical property of medical adhesivewill be enhanced under the incasement of the microspheres quantity. Interaction betweenmicrospheres and medical adhesive is relatively close which obtains good compatibility. Therelease experiment in vitro showed that:5-fluorouracil could release slowly under thedegradation of medical adhesive, the rate of releasing climbs with the increasing amount ofmicrospheres in medical adhesive and decreases with the decreasing of simulated pH in bodyfluid.
