Peptide chemistry has shown renewed spirit due to its development of peptidicscience and applications in the field of materials and life science. As one of the mostimportant functional peptides, the peptides with specific affinities to metal ions haveattracted significant interest because of their advantages in metal ion reduction andnanomaterials preparation. The nanomaterials fabricated from this kind of peptidesalways show low toxicity, good biocompatibility, and biodegradability. In addition,they show potentials for drug delivery, biological imaging, and data storage and soon. Therefore, it is very important to design and synthesize different kinds ofpeptides with high affinity. Oligopeptide with sequence AYSSGAPPMPPF has beenproven to show specific affinities to noble metal ions and possess remarkableadvantages in fabricating nanomaterials. In present thesis, we developed a novelmethodology to synthesize this water soluble peptide with high yield and purity. Tothe best of our knowledge, this is the first report referring to synthesis ofAYSSGAPPMPPF in solution. It started from single serine, followed by selectiveprotection or deprotection of the functional group and amide coupling reaction toafford the target product AYSSGAPPMPPF. Both the product and intermediate areunambiguously characterized by1H NMR and MS. The molecular brushes withAYSSGAPPMPPF as side chains were prepared and characterized with GPC and1HNMR spectroscopy. Circular dichroism (CD) spectroscopy was applied to investigatethe secondary structures of oligopeptides and the corresponding polymers.
