The rapid development of the modern industrial economy increases the developmentand utilization of mineral resources, followed by the formation of Tailings. Long-termaccumulation of tailings and its leakage poses a threat to the safety of the surroundingecological environment. Recent research has focused on the tailing dam safety and itsrelevant preventive measures at home and abroad, however, less are the leakage monitoringand pollution assessment of the tailings, also research on the toxic effect of tailing pondsurrounding water quality on biological system is rarely reported. In this study, in order toclarify the northern tailing pond surrounding water pollution, we combined thephysiochemical method and biological monitoring to further analyze the tailings waterleakage. As a result, we determined the physiological toxic effect of water pollution on itssurrounding environment, which acts as a warning to the safety and living of surroundingresidents, and provides scientific basis for the government to control the leakage pollutioncaused by tailing poond.Our study takes a northern tailing pond as the research subject. First, we analyzedtailing pond surrounding water quality using both physiochemical and biological methods.Taken corn and loach as the main experimental materials, we also analyzed the toxiceffects of the seepage stress on these living organisms at morphological, physiological andcellular levels. Because of the severe and wide fluoride pollution in the surroundinggroundwater, we further studied the toxic stess under single pollution factor of fluorine.Though MSAP (methylation sensitive amplification polymorphism) and gene cloningmethod, we also analyzed the epigenetic changes on loach at molecular levels, especiallythe methylation, pattern and sites of recombinant DNA as well as methylation-related genesequences.According to the experiments, some conclusions can be drawn as follows.(1) The monitoring results for water quality of the tailing pond and the tailings seepage show that each measurement indicators has exceeded the standard level. Comparing thedetection results of underground water from surrounding area with the detection resultsover the years, the research shows that the trend of water pollution has aggravated atpresent. Moreover, the F value calculated by Nemerow index in each sampling point alsoshows that the pollution degree of underground water is increased gradually with thedecreasing distance from tailing pond.(2) Under the stress of the tailing seepage, the germination rate and the germ’s length ofCorn seed present an inhibiting trend with the increasing concentration of seepage, whichleads to the pondage of MaizeSeeding cell, whilst MDA content and SOD activity forphysiological indexes of maize also emerge a resistance characteristic. However, the SODactivity in liver and kidney tissue of Loach does not show an obvious change.(3)Under the stress of fluorine pollution, the content of fluorine in corn leavesaccumulates increasingly with the increase of fluorine concentration, which makesgermination index of corn seed represent a decreasing trend. Meanwhile, MDA content inmaize seeding leaves is also increasing and SOD activity displays firstly increased thendecreased tendency. The changing trend of MDA content and the SOD activity in liver andkidney tissue of Loach is similar to Corn, with the exception that SOD activity has adeclining tend significantly in concentration which higher than50mg/L.(4) This study adopts methylation-sensitive amplified polymorphism (MSAP)technology to study epigenetic information such as methylation level,the result shows thatthere is no obvious changes rule in methylation level with concentration of fluorinestress.Analysis results about Methylation characteristics of DNA sequence, and the loachpolymorphism of under different fluorine concentration stress methylation modificationsite recycle, cloning, finally successfully separated the118methylation site mutation ofDNA sequence.BLAST analysis showed that25methylation modification sites retrieval tothe homologous sequences exist;This study analysis the clone sequences characteristicabout the CpG dinucleotide repeat sequence of DNA methylation sequences.Presumably loach DNA genome “CpG” dinucleotide clusters concentration area is a potential site ofDNA cytosine methylation modification.This result is similar to the opinion of Jiao J Nwith analysis about tobacco segments.In conclusion, analyse of the quality of tailings surrounding underground watershowed that there existed serious pollution of tailings seepage.Taken both plants andanimals as our research subject, combined the results from morphology, physiology andcell level, we basically determined the toxic effects of tailings seepage on living creatures,which threatens the growth and development of both plants and animals. Single pollutionfactor of fluorine not only inhibits plant growth but also influences DNA genetic stabilityat molecular level. Therefore, we’ve made the suggestion that relevant governmentdepartments shouldpay special attention on thetailings seepage condition.
