The corn syrup is the main by-product of corn starch processing, mainly composedis soluble protein of corn and its degradation products (such as peptides, amino acids,etc.), in addition also contains lactic acid, calcium magnesium salt plant, soluble sugar,etc. Corn starch is rich in nutrients, but at present, corn pulp of some companies is notgood used, even direct emissions cause environmental pollution.The purpose of this research was to depth digging the excavation use value of cornsyrup, studied mainly corn soaking process, the fermentation process of Aspergillusniger phytase and phytase separation and purification process, corn steep liquor drypowder, calcium phytate, inositol preparation and fermentation applications,experimental results are as follows:1. Research soaking process, to prepare corn syrup dry powder. The best sockingconditions for static method:60h,50°C, and the sulfite comcentration of0.2%. Theoptimum processing for multi-tank series countercurrent immersion method:8cansseries, once every6h inverted cans, soak for a total time of48h, immersion end gotcorn soaking water pH is4.8, total sugar content is0.23mg/L, protein conten is0.26mg/L. Corn starch powder moisture content is3%, protein content is45%, reducingsugar content is4.6%, total phosphorus content is1.6%.2. Research of the fermentation process of Aspergillus niger for phytase andphytase separation and purification. The optimal medium of the Aspergillus niger liquidfermentation acidic phytase consisted of5%soluble starch,5%bean pulp,0.9%KCl,0.1%FeSO4·7H2O,0.9%MgSO4·7H2O,0.5%MnSO4. The effect of the factor is thefollowing order: MgSO4·7H2O, soluble starch, bean pulp, MnSO4, FeSO4·7H2O, KCl.The optimal fermentation condition was fermentation temperature30°C, the initial pH5.5, the shaking speed220r/min and the fermentation time60h, got the acidic phytaseenzyme activity reached20U. The phytase separation and purification conditions:717anion exchange resin, the sample buffer pH5.5, the eluent was0.6mol/L Nacl, got ahigh recovery rate was98%. The molecular weight of the phytase is about45KD, thebest temperature of enzyme activity is55°C, the best pH value is5.5, with thermalstability and pH atability.3. The study corn syrup production of calcium phytate and phytase production ofinositol process. Calculated through the yield of the phytin got the content is0.01~0.02g/mL in corn water, the content is0.075~0.09g/mL in corn starch. The process ofinositol preparation:30min,50°C, the amount of the enzyme5%, pH5.5,180r/min.Established reversed-phase high-performance liquid chromatography (HPLC) method todetect inositol content.4. Research of corn starch and its by-products in the application of themicroorganism fermentation. Corn starch powder used in conjunction with yeastpowder than used alone yeast is more advantageous to the growth of E.coli. The best accelerator concentration of S. cerevisiae fermentation is0.6%phytin concentration.Inositol has a certain influence on the fermentation of S. cereivisiae and ethanoltolerance,150mg/L of inositol content is more suited to the fermentation of S.cerevisiae, under this condition, the S. cervisiae tolerated12%alcohol content.
