China is the major producing countries and the only exporting country, the share of the world jujube productionis more than90%. Undoubtedly the jujube polysaccharide is the most abundant and obvious medical effects.It most likely to achieve industrial production and application of jujube deep processing products. Studies have shown that jujube polysaccharide with significant anticomplement activity and promote lymphocyte proliferation, antioxidant, antiaging, antiradiation, enhance antibody immunity has an important role in the natural active ingredients of jujube.At present,anticoagulant medicine was mostly used for clinic treatment was heparin,which had some side-effects, such as causing platelet disease, hemorrhage disease, hyperkalemia and so on. So it was essential to develop new type of medicine or functional food.The experiments mainly reached following results:1. The results showed that Jujube crude polysaccharide had the anticoagulant function, which could effectively prolong activated partial thromboplastin time (APTT) of human plasma. But for prothrombin time (PT) and thrombin time (TT) had no significant effect, indicating that the anticoagulation function of Jujube Polysaccharide mainly through the intrinsic coagulation system.2. The anticoagulant activity of different jujube varieties crude polysaccharide have significantly different between13varieties of anticoagulant activity, which Lingbao jujube anticoagulant activity is most significant. In addition, there is no obvious correlation between the strength anticoagulant of activity jujube polysaccharide and neutral polysaccharides, acidic polysaccharides, protein ratio.3. The Anticoagulant activity of Jujube crude polysaccharide is significantly different by four extraction method,(water formulation, the ultrasonic method, microwave method and ultrasonic auxiliary enzymatic) and four drying methods (hot air drying, microwave drying, vacuum drying, vacuum freeze drying). Ultrasonic extraction and hot air drying and vacuum freeze drying could effectively protect the anticoagulant function of jujube crude polysaccharide.4. Ultrasound supplementary technology was used in this experiment in order to increase the yield and protect the anticoagulant function of jujube crude polysaccharide. Single factor experiment were arranged to analyze the effects of factors such as extraction temperature, ultrasonic power, ultrasonic time, pH value and ratio of liquid to solid on the extraction of jujube crude polysaccharide and their anticoagulant activities. Based on the response surface analysis with the Design-Expert software program, the extraction condition was optimized. The results showed that optimum extraction conditions were temperature69"C, pH7.15, ultrasonic power80W, ultrasonic time30min, the ratio of liquid to solid1:10. The result could effectively protect the anticoagulant function of jujube crude polysaccharide, besides the highest yield.5. The anticoagulant activity of Jujube Crude polysaccharide was enhanced throngh deproteinization and AB-8macroporous resin chromatography. Three components of jujube polysaccharides(JP Ⅰ, JP11, JPⅢ) were purified from by DEAE-52cellulose column. JPI was the main component of the jujube polysaccharide and the anticoagulant activity was significantly higher than the other two components, molecular weight of29922D. The anticoagulant activity between JPII and JP III was not significantly.Their molecular weights were41686D and54200D respectively.6. The purity of JPI, JPII, JP III were verified by Sephadex G-100, UV spectrophotometry and frozen-thawing analysis.7. IR spectrum analysis showed that two components of Jujube polysaccharides all have the typical character of polysaccharides JPIand JPII, appeared absorption peak at894cm-1and892cm-1respectively, they contained P-glycosidic bond, containing acidic polysaccharides. JPII has relatively high-COOH group and also has an absorption peak of the mannose.8. The jujube polysaccharide JPI, JP II carried sulfuric acid modification in vitro anticoagulant activity was significantly enhanced.
