Jujube pigment,extracted from jujube residues during jujube juice processing,is a water-soluble pigment containing polyphenols and flavonoids.It is an ideal natural pigment resource and has an ability to clear the DPPH,ABTS+,OH,and O2-.Besides,it has a certain inhibitory effect on lipid peroxidation and can improve the damage of vascular endothelial cells induced by H2O2.Jujube Polysaccharide is a mixed polysaccharide extracted from the fruits of the Rhamnaceae plant and can eliminate OH,O2- and H2O2.It functions good antioxidant activity and anti-aging,and also can enrich the blood,protect liver,and enhance immune function by directly acting on immune cells.Due to the complexity of natural food ingredients,antioxidants in natural food extracts are often not a single ingredient,but a mixture of multiple ingredients.Synergistic or antagonistic effects may occur between natural antioxidants.Therefore,research on the synergies between them are of great significance for enhancing antioxidants.At present,most of the studies in the literature have reported the antioxidant activity of single factor in natural plants.There are also many studies on the synergy between antioxidants from different sources.However,there are few reports on the interactions between different factors from the same source.Therefore,this experiment aims at studying the synergistic effect of jujube pigment and polysaccharide with antioxidant activity in vitro and anti-lipid peroxidation on relieving fatigue and enhancing immune function of Mice.The synergistic effect of red jujube pigment and jujube polysaccharide has been discussed so as to provide the theoretical foundation for industrialized development of red dates,as well as the development and utilization of red jujube pigment and polysaccharide.Additionally,it's also expected to provide the theoretical basis for the joint use of natural antioxidants and for diseases prevention,like diabetes,cardiovascular and cerebrovascular,and so on.The results are as follows:(1)The synergistic effect in vitro antioxidant activity of jujube pigment and polysaccharide has been studied.The results show that the coordination of synergistic jujube pigment and polysaccharide can effectively remove DPPH,ABTS+,OH,O2-,H2O2 and NO2-,which also has good reducing power and total antioxidant capacity.Its scavenging ability gradually increases along with the increase of concentrations.What is more,the scavenging ability of the combined jujube pigment and polysaccharide is stronger than the jujube pigment or the polysaccharide used alone.From the radiometric analysis,we can see that the effects of the jujube pigment and polysaccharides are on the left side of the additive line and the 95%confidence limit.The experiment value EC50,mix is less than the theoretical value EC50,add(p<0.05),and the interaction index(?)is less than 1.The synergistic rate of eliminating DPPH,ABTS+,-OH,O2-,H2O2 and NO2-are respectively 53.53%?61.64%?85.73%?65.34%?66.06%and 80.88%.The synergy rate of reducing power and total antioxidant capacity are 87.04%and 70.51%,which reflects the fact that the combination of red pigment and polysaccharide obtains the anti-oxidation synergistic effect.(2)The synergistic anti-lipid peroxidation effect of jujube pigment and polysaccharide has been studied.The results show that red jujube pigment and polysaccharide can effectively enhance the lipid peroxidation capacity of soybean oil,yolk lipoprotein,linoleic acid,mouse liver tissue and the lipid peroxidation system of mouse red blood cell.The inhibitory ability gradually increases along with the increase of concentration.The anti-lipid peroxidation ability of joint use of red jujube pigment and polysaccharide is stronger than the use of red jujube pigment or polysaccharide alone.From the radiometric analysis,we can see that the effects of the jujube pigment and polysaccharides are on the left side of the inhibition of soybean oil,Fe2+ induced yolk lipoprotein,linoleic acid,Fe2+ catalyzed linoleic acid,spontaneous liver tissue in mice,H2O2 induced liver tissue in mice,H2O2+Fe2+ combination induced liver issue in mice,H2O2-induced erythrocyte membrane lipid peroxidation and H2O2-induced red blood cell hemolysis are 69.77%,78.85%,88.87%,36.28%,33.43%,50.44%,56.70%,36.02%,and 74.57%,respectively.It indicates that red jujube pigment and jujube polysaccharide have synergistic effect on lipid peroxidation.(3)The animal swimming test has been applied to study the red jujube pigment and polysaccharide on relieving fatigue in mice.The test results show that the combination of red jujube pigment and jujube polysaccharide has no significant effect on weight of mice,but can significantly prolong the exhausting swimming time of the mice,and increase the blood glucose,hepatic glycogen and muscle glycogen content after exercise.Meanwhile,the red jujube pigment and polysaccharide can also reduce the content of blood lactate,serum urea nitrogen,and malonaldehyde,and decrease the activity of creatine kinase,lactate dehydrogenase and alanine aminotransferase in blood and in turn increase the activity of catalase and superoxide dismutase.The effect for red jujube pigment and polysaccharide synergistically relieving the fatigue of mice is stronger than the same dose of red jujube pigment or polysaccharide used alone,which shows that the combination of red jujube pigment and jujube polysaccharide has a good help in relieving physical fatigue.(4)An immunocompromised mouse model of intraperitoneal injection of cyclophosphamide has been established to study the synergistic effect of red jujube pigment and polysaccharide on immune function in mice.The test results show that the index of immune organs,the clearance index of carbon particles,the serum hemolysin content of mice,and the transformation ability of spleen lymphocytes of mice are all increased after red dates pigment and jujube polysaccharides being compounded.The immune function of rats is stronger than that of the same dose of red jujube pigment or polysaccharide used alone,which also verifies the fact that the jujube pigment and polysaccharide have a synergistic effect.They have the function of synergistically enhancing the cellular immune function of mice.
