| The extraction of Trichosanthes polysaccharide, Trichosanthes skin beverage,physicochemical properties, flavor compounds, amino acid composition and antioxidantactivity were studied and results were as follows:The optimal extraction conditions of material-to-liquid ratio, extraction time andmicrowave power were1︰40,15min and450W respectively. Under these extractionconditions, the values of polysaccharides yield reached to2.52%. The antioxidant activityof Trichosanthes kirilowii skin polysaccharide was evaluated by total reducing power,hydroxyl radical scavenging assay and NO2-radical scavenging assay. The reducing power,hydroxyl radical scavenging rate and NO2-radical scavenging rate of the1.2g/mL ofpolysaccharide polysaccharide were0.54(OD),72.8%and35.6%.The best prescription of Trichosanthes skin beverage is raw Trichosanthes juicecontent9.77%, granulated sugar6.86%and citric acid0.05%. The best formula stabilizeris Arabia gum0.10%, CMC-Na0.10%and agar0.06%. Under the optimal condition,The results of sedimentation rate of Trichosanthes skin beverage was2.95%. Acute oraltoxicity test, micronucleus test and mouse sperm malformation rate experiment resultsshowed that, there was no significant difference between the experimental group andnegative control group (P>0.05). Meanwhile, no pathogenic microorganisms were found.Trichosanthes skin-orange compound beverage (TOB) was prepared with the air driedTrichosanthes skin and the Sunkist orange at the ratio of3︰40(w/w). The total contents ofthe phenols, flavonoids, dietary fiber and VC were285.82(μg GAE/100g),18.23mg/L,5.64g/L and108.76μg/mL respectively.16kinds of amino acids were detected in TOB,contents of arginine, cysteine and tyrosine were obviously higher than those of OB and TB.25kinds of flavoring substances were detected in TOB, mainly including the olefin, alkanesubstances and acids, small contents of ketone and aldehyde substances were also included.Contents of cyclohexane and aromadendrene were greatly higher than those of OB(P<0.05). The general acceptabilityof TOB (9.1points) was obviously higher than that ofOB and TB. During storage, the antioxidant activity of TOB was found to decrease.However, after60days of storage relative high scavenging ability of DPPH, ABTS andFRAP with values of1.66ã€3.05ã€2.44mM AA were detected.Trichosanthes skin-milk fermentation beverage (TMB) fermented by S. thermophilusand L.bulgaricus were prepared with different concentration of air dried Trichosanthes skin juice(10%ã€15%ã€20%ã€25%)and5%of milk. The number of lactic acid bacteria, pH,titratable acidity and the content of lactic acid of TMB performed no significant difference(P>0.05) to the control group. The firmness, consistency, cohesiveness and index ofviscosity of TMB were significantly decreased (P<0.05). With the increase of Trichosanthesskin juice content, contents of Asp, Val, Ile, Tyr, Phe and His in TMB were increasedgradually. The general acceptability of TMB was8.6points, which was obviously higherthan that of control beverage (P<0.05). The scavenging abilities in reletion toDPPH· and·OH, chelating ability in reletion to Fe2+of TMB were obviously higher thanthat of the control beverage (P<0.05).The ultrasonic assisted extraction method was selected to obtain the Trichosanthesseed oil and the yield was23.93%. The results of UV spectroscopy showed that263,273,283nm were the main UV absorption wavelength. Infrared spectroscopy showed that theTrichosanthes seed oil contains9c,11t,13c-CLNA. GC-MS analysis showed that9c,11t,13c-CLNA relative content of29.37%. Raw materials of the prepared blend oil weresunflower seed oil, soyabean oil and Trichosanthes seed oil. The optimal proportion of eachcomponent in blend oil was determined by MATLAB6.1with VE content as the highestindex. That’s500g of blend oil need to add sunflower seed oil227.57g, soybean oil209.28g and Trichosanthes seed oil63.15g. Trichosanthes seed blend oil exhibited higheststability under air condition, light condition and fried condition, when compared to otherfats. Acid value, iodine value and peroxide value of Trichosanthes seed blend oil weredetermined and it showed an acid value of0.34mg KOH/g, iodine value of123.7g I2/100gand peroxide value of7.2nmol/kg. |
