Phytic acid is a natural compound which can be widely found in corns,legumes and oil-seeds. It has attracted great attention by its application in foodindustry, environment protection and medical treatment depending on its uniquechelation with metals and natural anti-oxidization. Therefore, the exploitation ofphytic acid using our abundant plant resources has a broad prospect.Phytic acid extraction processing was studied from rice bran in thisexperiment. The extraction conditions of acidic solution were optimized, andcombined with membrane separation technology; the process of membraneseparation was optimized. Then static adsorption and elution of phytic acid bydifferent macroporous resins were studied so as to choose the best from them toresearch dynamical adsorption and elution. The structure of sample phytic acidmade in this experiment was also verificated and validated by the method ofInfra-red and NMR spectroscopy.The main conclusions of the experiment were as follows:(1)The process of extraction was optimized by orthogonal experiment on thebasis of unifactor experiments with phytic acid extraction rate as the index. Theoptimal conditions were:extraction agents was HCl, extraction temperature40℃,extraction time2.5h, extraction pH3.0, the ratio of liquid to material10:1, underwhich the extraction rate of phytin was12.08%, the purity of phytin was78.12%.(2)Analysing the treatment effects of three kinds of membrane RF1, RF2andRF3to pickle liquor, and the process of membrane separation was optimized.Results indicated that the membrane RF3was better than other membrane and theoptimal conditions were: time20min and diafiltrations.The penetration of phyticacid was90.36%, the protein retention ratio was83.18%.(3)The performances of absorption and elution by kinds of resins to the phyticacid form rice bran were studied.D301resin was chosen to explore the staticadsorption condition. Based on static method, the influencing factors such asequilibrium time, temperature, pH and the type and concentration of eluant werestudied. The results showed that the optimal conditions were: adsorption temperature25℃,time2h, pH3and elution concentration of NaOH1mol/L.(4)Macroporous resin D301in the dynamic adsorption and elutionperformances of phytic acid were studied. Kinds of ion, resin height, sampleconcentration, sample flow rate on the impact of resin adsorption and eluantconcentration and flow rate on the impact of resin elution were examined. Theresults showed that: the resin of Cl D315, the diameter to height ratio1:10, thesample concentration3mg/mL and the sample flow rate1.0mL/min were better foradsorption; concentration of NaOH1.0mol/L and the eluting flow rate2.0mL/minwere better for elution.(5)After purifying,the structure of phytic acid was determined by means ofInfra-red and NMR spectroscopy, and the quality was tested.The IR spectrum andNRM spectrum of phytic acid in this experiment were contrasted to standardspectrum, and what was extracted was confirmed to be phytic acid. Finally, thecrystal structure of phytic acid was determined by means of XRD.
