Flos Puerariae and Hovenia dulcis Thunb were main raw materials in this experimentation. The main chemical ingredients of Flos Puerariae were flavonoids, triterpenoid saponins, coumarins, sterols, volatile oil, alkaloids and essential amino acids and inorganic salts that can not be synthesized by human body. Hovenia dulcis Thunb is buckthorn plants seed, it is considered to be Yin fluid possessing, runchang purge and alleviating wine, etc. by Chinese medicine. Both of them are traditional anti-hangover recipe and as main materials in this experimentation. High efficient ball milling technology was applied to prepare Sober-up chewable tablets absorbed and utilized completely by human body. Sober-up chewable tablets will be mainly used to alleviate symptoms of drunk, sober time, recuperate chronic diseases caused by long-term heavy drinking. The detailed results are as follows:Firstly, the properties of Flos Puerariae and Hovenia dulcis Thunb powder were studied. Based on the total material dissolution, total flavonoids dissolution and alcohol dehydrogenase (ADH) activity in vitro, rutin was as standard substance and ultraviolet visible spectrophotometry was used to determined chromogenic reaction to calculate the the total material dissolution. The result showed that the total material instantaneous dissolution of Flos Puerariae II powder is8.6times higher than the untreated samples. In the subsequent determinations of six time point, the total material dissolution was higher than the other groups, and its rate tended to stability after20min. The change tendency of different particle sizes of Hovenia dulcis Thunb was similar to that of different particle sizes of Flos Puerariae. The instantaneous dissolution of grade II powder was13.5times higher than that of untreated plants, while in the determination of six time point, the total material dissolution was higher than the other groups. In terms of the determination of total flavonoids dissolution,2.67mg/g of untreated Flos Puerariae was increased to9.75mg/g of grade Ⅱ powder. Compared with12.21mg/g flavonoids dissolution extracted by80%ethanol3times, flavonoids dissolution of grade Ⅱ powder was increased by57.98%. Similarly, flavonoids dissolution of Hovenia dulcis Thunb was increased by64%. By analyzing effects of different particle sizes of Flos Puerariae and Hovenia dulcis Thunb on the activity of ADH, it suggested that both of them could activate alcohol dehydrogenase in vitro, while the grade II powder obtained by using efficient milling technology was better than other powders, its activation rate reached37.8%and2.9%, respectively. Secondly, HPLC/HPLC-MS2was applied to study isolation and identification of flavonoids obtained from Flos Puerariae. a single peak was obtained through HPLC technology after Flos Puerariae samples were pre-processed, and then HPLC-MS2was used to analyse its structure. According to second-order MS spectrum of positive ions, the function factor possessing hangover effect in Flos Puerariae was determine to be Tectoridin.Finally, grade Ⅱ powder of Flos Puerariae and Hovenia dulcis Thunb was selected as the main raw material to produce Sober-up chewable tablet. As for producing tablet technique, five ingredients, such as sweeteners, acidulant, disintegrants, fillers and wetting agents, were choosed to evaluate appearance and taste of the tablet. The quality index of tablet was as standard for evaluation, the optimum proportions of the tablet were34.5%sweeteners,0.75%sour agents,16.7%disintegrating agents,14.8%fillersagents and0.36mL/g wetting agent by using comprehensive fuzzy evaluation method. Texture Analyzer with2mm Cylinder Probe (P/2) probes was applied to compare the differences between the chewable tablets and toffee. The results showed that hardness of chewable tablet was2114.78g. By investigating flavonoid dissolution of chewable tablet and alcohol dehydrogenase (ADH) activity in vitro, the results showed that flavonoid dissolution was2.7mg/g, ADH activation effect was positive and the activation rate was up to36.7%.
