Puerariae flos is a traditional Chinese herbal medicine,tectoridin and tectorigenin extracted from Puerariae flos have a variety of pharmacological activity.And the activity of tectorigenin is higher than that of tectoridin.In this paper,the process of extraction of tectorigenin from Puerariae flos was optimized with Puerariae flos as the raw material.The antioxidant activity and inhibition of tyrosinase activity were evaluated.The application of tectoridin and tectorigenin in the mask was studied.The main results indicated as follows:1.The tectorigenin was exstracted from Puerariae flos by using acid hydrolysis and ethanol extraction.On the basis of single factor(acid hydrolysis time,acid hydrolysis temperature,hydrochloric acid concentration and solid-liquid ratio)experiment,optimize orthogonally designed experiment of three factors and three levels to get the optimal process conditions:acid hydrolysis time 6h,acid hydrolysis temperature 90?,hydrochloric acid concentration 1mol/L and the ratio of solid-liquid 1:40.In this condition,the yield of tectorigenin was 1.89%,and the purity was about 60.78%.2.The evaluation of antioxidant activity of tectoridin and tectorigenin in vitro studies.The experiment of reducing power show that the antioxidant activity of tectorigenin was higher than that of tectoridin in the range of 0 mg/mL?0.15 mg/mL.The experiment of activity of scavenging DPPH and ABTS+free radicals showed that the antioxidant activity of tectorigenin was higher than that of tectoridin in the range of 0.01 mg/mL?0.2 mg/mL.The results showed that the method of directly extracting tectorigenin by acid hydrolysis and ethanol extraction can maintain higher antioxidant activity.3.The mixed solvent of propylene glycol and ethyl acetate(V(propylene glycol):V(ethyl acetate)=9:1)was used as the dissolution reagent of the tectoridin and tectorigenin for analyzing the inhibiting effect of tectoridin and tectorigenin on tyrosinase.Experimental results showed that the inhibitory effect of tectorigenin on tyrosinase was higher than that of tectoridin,but the inhibitory effect of tectorigenin on tyrosinase was reduced in high concentration.That's a two-way effect.4.The optimum formulations of two different types of mask were determined by using the physical and chemical properties of the product,sensory properties and rheological properties as the evaluation indicators.The optimum formulation of clay facial masks:2 parts of tourmaline powder,2 parts of bentonite,2 parts of kaolin,4 parts of glycerin,1 part of water and 1 part of tectoridin(or tectorigenin)solution;the optimum formulation of gel type facial masks:8 parts of hydrothermal mask matrix basic liquid of facial masks,1 part of water and 1 part of the mixed solution of 0.2%xanthan gum and tectoridin(or tectorigenin).Antioxidant whitening effect evaluation and skin sensitivity test results showed that two different types of mask both had a good effect.
