Choerospondias Axillaries which has high medicinal and edible value is a kind of deciduous shrub or small tree of Zizithus Mill of Rhamnaceae.The study evaluated the antioxidant effect of choerospondias axillaries extract (CAE) in drosophila melanogaster, so that we can provide the scientific basis for the further research and development of Choerospondias Axillaries.The purpose of this part was studies on anti-aging of CAE in drosophila melanogaster in the normal group.The results which concluded via the survival test of fruit flies and biochemical test showed the median lethal time, maximum lifespan and mean lifespan of the fruit flies treated with different doses (1、5、10、15mg/mL)of CAE at25d and40d was higher than the control group. With the doses of CAE increased in the experiment, the activities of CAT, T-SOD, CuZn-SOD was increased while the content of maleic diadehyde (MDA) was reduced.The activities of antioxidant enzymes in fruit flies at25d was higher than they were at40d, the content of MDA at25d was lower than it was at40d.Thus, the research indicates CAE has obvious capacity of antioxidation.The purpose of the second part was studies on anti-aging of CAE in drosophila melanogaster damaged by UV.The best dose of UV radiation was selected while the basal diet were supplemented with0,1,5,10or15mg/ml CAE. With the doses of CAE increased in the experiment, all of mean lifespan, the maximum lifespan and median lethal time were longer than model group in Drosophila melanogaster, and the activities of CAT, T-SOD, CuZn-SOD in vivo were increased, while the content of MDA was reduced.The purpose of the third part was studies on anti-aging of CAE in drosophila melanogaster damaged by fat. The model of hyperlipidemia flies was established and different dosages of CAE were fed to flies. The flies fed on CAE are much more macrobian compared with the model groups. T-SOD, CuZn-SOD, CAT activities were improved significantly and the content of MDA reduced with dose enhancement. The otherness is significantly between groups.The purpose of the fourth part was studies on anti-aging of CAE in drosophila melanogaster damaged by H2O2and Paraquat.The model of normal flies in acute experiment was established and different dosages of CAE was fed to flies. And then the flies’hydroxyl radical and superoxide radical anion was generated by H2O2and Paraquat. The result was that the mean lifespan, the maximum lifespan and median lethal time was all prolonger than the control group. Meanwhile, the three index of high-dosage groups was longer than low-dosage.The purpose of the fifth part was studies on gene expression of antioxidant enzymes of CAE in Drosophila melanogaster. RNA was isolated by the Trizol method. And the results showed the purity tested by spectrophotometer and the integrity tested by Agarose gel electrophoresis technology of the RNA in flies was good. Then it was reversed transcriptase into cDNA and conducted on PCR amplification by SYBR Green fluorescence quantitative method.The results showed that gene expression of the CAT, CuZn-SOD, MnSOD mRNA were increased compared with the control.CAE which can be used as raw material of health care products has capacity of antioxidant and senescence-resistance.
