Purple sweet potato,also known as black potato,is rich in anthocyanins and other active ingredients with high medicinal and edible value.In this paper,Drosophila melanogaster and Caenorhabditis elegans were choosed as experimental model organisms.PSPE in vivo antioxidant activity were studied in order to evaluate the antioxidant capacity and effect of aging regulation of PSPE.The preliminary study of the molecular mechanism of its action and further exploration of PSPE antioxidant mechanisms were also been explored,which will provide some guidance for the development of related products.In this paper,LC-MS/MS was as the main way to make the qualitative and quantitative analysis of active ingredient with PSPE.The drosophila and nematodes were fed with different dose(Drosophila melanogaster 0,0.5,2.0,5.0mg/mL;Caenorhabditis elegans 0,70,140,280?g/mL)of PSPE.The two model organisms were as to study impacts of PSPE on the lifespan,the acute stress tolerance,the antioxidant enzyme activity,the changes in lipid peroxidation product malondialdehyde(MDA)and antioxidant-related genes mRNA expression levels.This paper found that PSPE can significantly prolong the lifespan of drosophila,improve the ability of crawling,enhance the activity of antioxidant enzymes containing SOD and CAT,raise relevant anti-aging gene expression level and inhibit the accumulation of lipid peroxidation products and.While the same as the activity and lifespan of drosophila in challenge test of paraquat and hydrogen peroxide.It also had a good protective effect on oxidative damage of high fat and UV damage of drosophila.Thus,the conclusion indicates PSPE has the obvious capacity of antioxidant.It was also shown that the lifespan of C.elegans were prolonged obviously by PSPE addition.The antioxidant enzymes activity of SOD were increased with the regulation of the expression levels of genes related to aging.The acute results including the heat stress experiment,UV radiation stress experiment,hydrogen peroxide oxidative stress experiment,juglone oxidative stress experiment and paraquat oxidative stress challenge test show that the flies treated by PSPE with the longer Survival time compared to that of the control group.Besides the lipofuscin autofluorescence level in vivo of C.elegans significantly decreased and it was dose-dependent effect to the concentration of PSPE.This study showed that PSPE can effectively delay aging of model organisms,regulate the mRNA expression levels of the relevant endogenous antioxidant genes,enhance the activities of antioxidant enzymes,improve antioxidant capacity and the ability to scaveng free radicals.In conclusion,PSPE has good effects of antioxidant activity and delaying senescence.
