In this paper, a set of improved vibrio selective differential medium with the research object of vibrio damsela, vibrio mimicus, vibrio cincinnatiensis and vibrio vulnificus was devoloped. And the specificity, sensitivity and accuracy of improved vibrio selective differential medium were evaluated and compared with those of the initially developed vibrio selective differential medium. It was hoped that the differentiating effect, testing cost, etc. of improved vibrio selective differential medium had been improved.The researches carried out in this paper would be described as follows:1. Study on the improved vibrio damsela selective differential medium (imVdSDM)An improved Vibrio damsela selective differential medium (imVdSDM) was developed according to the initially developed Vibrio damsela selective differential medium (VdSDM) combined with the special enzyme system, metabolic characteristics, the tolerance of specific antimicrobial substance and other biochemical characteristics of V. damsela. These special biochemical characteristics were selected according to relevant information and experimental studies. The testing results showed that, after incubated for24h, V. damsela formed purple colonies on imVdSDM, and other species of bacteria could not form colonies or form yellow colonies on imVdSDM. The detection limit and positive rate of imVdSDM were respectively101cfu/mL and100%. In addition, the specificity, sensitivity, accuracy and the performance in the detection of V. damsela from natural samples of imVdSDM were good.2. Study on the improved vibrio mimicus selective differential medium (imVmSDM)An improved Vibrio mimicus selective differential medium (imVmSDM) was developed according to the initially developed Vibrio mimicus selective differential medium (VmSDM) combined with the special enzyme system, metabolic characteristics, the tolerance of specific antimicrobial substance and other biochemical characteristics of V mimicus. These special biochemical characteristics were selected according to relevant information and experimental studies. The testing results showed that, after incubated for18h, V. mimicus formed purple colonies on imVmSDM, and other species of bacteria could not form colonies or form yellow colonies on imVmSDM. The detection limit and positive rate of imVmSDM were respectively101cfu/mL and100%. The specificity, sensitivity, accuracy and the performance in the detection of V. mimicus from natural samples of imVmSDM were good. In addition, the cost of each plate reduced from about5.58yuan for VmSDM to about0.33yuan for imVmDM, with which testing cost was effectively cut down.3. Study on the improved vibrio cincinnatiensis selective differential medium (imVciSDM)An improved Vibrio cincinnatiensis selective differential medium (imVciSDM) was developed according to the initially developed Vibrio cincinnatiensis selective differential medium (VciSDM) combined with the special enzyme system, metabolic characteristics, the tolerance of specific antimicrobial substance and other biochemical characteristics of V. cincinnatiensis. These special biochemical characteristics were selected according to relevant information and experimental studies. The testing results showed that, after incubated for24h, V. cincinnatiensis formed yellow colonies on imVciSDM, and other species of bacteria could not form colonies or form green or blue-green colonies on imVmSDM. The detection limit and positive rate of imVciSDM were respectively101cfu/mL and100%. The specificity, sensitivity, accuracy and the performance in the detection of V. cincinnatiensis from natural samples of imVciSDM were better compared to VciSDM and TCBS. 4. Study on the improved vibrio vulnificus selective differential medium (imVvSDM)An improved Vibrio vulnificus selective differential medium (imVvSDM) was developed according to the initially developed Vibrio vulnificus selective differential medium (VvSDM) of which the cost is relatively high. And in order to find a better reaction substrate for imVvSDM, the special enzyme system and metabolic characteristics of V. vulnificus were researched. The cost of each plate reduced from about5.72yuan for VvSDM to about3.36yuan for imVmDM, to some extent reduced the testing cost. In addition, the evaluation test showed that the identification effect of imVvSDM was as good as VvSDM.The four improved vibrio selective differential medium had improved the specificity and to some extent reduced testing cost of the relevant four initially developed vibrio selective differential medium. And they would be respectively useful for the rapid detection of V. damsela, V. mimicus, V. cincinnatiensis and V. vulnificus in aquatic products.
