In this paper, water samples obtained from mines in northern Anhui, from whichsulfate-reducing and methanogenic bacteria were cultured and isolated, detected aproduction of both H2S and CH4. Their morphologies were observed, and DNAidentification of16S rRNA amplification were used to identify the strains.Theexperimental study on the effects of temperature and pH on the gas production ofMethanogens.After verifying mine groundwater samples contain sulfate-reducing bacteria andmethanogenic bacteria, simulated coal groundwater in a closed environment, andStudied on the sulfate-reducing and methanogenic bacteria and their Symbiosis.Thisstudy selected typical filling aquifer water from mine water in closed condition,studied the specific quantitative indicators of microbial alternative process,oxidationreduction potential, water-soluble ions, sediment, then based on the detection ofsulfate reducing bacteria and methanogenic bacteria in the output low redoxconditions, draw a new clear ecidence that two kinds of bacteria can be symbiotic.However due to the higher concentration of SO42-,before the sulfate-reducing bacteriabean to play a part, methanogenic bacteria’s action was inhibited.The experimentmeasured that under the role of sulphate reducing bacteria, SO42-concentrationdecreased, then afer most of the SO42-is reduced, it began to produce CH4, thusproved that methanogenic bacteria began to work.Select the first coal mine of Liuqiao, the second coal mine of Liuqiao and Juji coalmine, Panbei coal mine, Panyi coal mine to conduct case study.Panbei coal mine andPanyi coal mine are ahigh-gas coal mine, while the first coal mine of Liuqiao, thesecond coal mine of Liuqiao and Juji coal mine belonging to low gas coalmine.Through analyzing water quality data of the mine aquifer, draw the Piperdiagram.From the Piper diagram it can be seen that the content of SO42-in Panbei coalmine and Paner coal mine is extremely low, because the collection of the watersample got the smell of rotten egg, that shows it contains large amount of H2S gas,which indicates that this area of sulfate reducing bacteria and sulfate is very strong.High SO42-content in the first coal mine of Liuqiao, the second coal mine of Liuqiaoand Juji coal mine. Therefore by analysis the mine case, it accord with theexperimental results in Chapter IV. It can come to the conclusion that thesulfate-reducing bacteria and methane-producing bacteria in coal groundwater can be symbiotic, and at higher SO42-concentrations, it will inhibit the activity ofmethanogenic bacteria.
